摘 要:目的综述类器官模型的新型技术及其在结直肠癌耐药性中的不同应用现状。方法以“类器官、结直肠癌、结直肠癌治疗、结直肠癌耐药性”为中文关键词,以“organoids、colorectal cancer、treatment of colorectal cancer、drug resistance in colorectal cancer”为英文关键词,检索PubMed和中国知网数据库自建库至2024-08-31发表的相关文献。纳入标准:类器官发展;类器官模型的构建;结直肠癌的治疗;结直肠癌耐药性;类器官与结直肠癌耐药性。排除标准:会议摘要及中文学位论文等非正式发表的文献。依据纳入和排除标准,最终纳入64篇文献进行分析。结果类器官是在干细胞技术基础上,通过器官前体细胞的自组织过程发展而来的体外三维培养物,由于类器官技术能够复制来源组织基因表达谱及相应器官的关键特征和功能,因此在组织结构上与在体组织高度相似。类器官在结直肠癌化疗、靶向治疗等耐药性研究中均取得了有效的进展,特别是利用类器官探索关键通路、关键靶点以及肿瘤微环境的改变所导致的耐药性。但是现有的类器官缺乏与间质细胞、免疫细胞之间的相互作用,不能反应在肿瘤转移过程中与各种器官组织的相互作用。结论类器官在结直肠癌对化疗、靶向治疗、免疫治疗耐药性的研究中取得了一定进展,但是类器官培养条件以及与器官间的相互作用仍然面临一定挑战。s and Chinese disertations.A total of 64 articles were ultimately included.Results Organoids are in vitro three-dimensional cultures developed through the self-organization process of organ precursor cells on the basis of stem cell technology,which are highly similar to in vivo tissues in terms of organization and structure due to their ability to replicate the gene expression profile of the source tissues as well as the key features and functions of the corresponding or-gans.Organoids have made effective progress in the study of drug resistance in both chemotherapy and targeted therapy for colo-rectal cancer,especially in the use of organoids to explore key pathways,key targets,and drug resistance due to altera-tions in the tumor microenvironment.However,existing organoids lack interactions with mesenchymal cells and immune cells,and can not respond to interactions with various organs and tissues during tumor metastasis.Conclusion Organoids have made some progress in the study of colorectal cancer resistance to chemotherapy,targeted therapy,and immunother-apy,but the culture conditions of organoids and their interactions with organs still face some challenges.
