摘 要:肾透明细胞癌是成人肾癌中最常见和最致命的泌尿系统癌症,其中PI3K/Akt信号通路的异常激活在肾透明细胞癌发生和发展中起重要作用。PTEN是PI3K/Akt信号通路的关键抑制基因,PI3K/Akt信号通路的异常激活是由PTEN基因功能下调或失活导致。因此了解肾透明细胞癌中PTEN基因功能下调或失活的作用机制和影响因素,对调控PI3K/Akt信号通路的异常激活至关重要。本文旨在总结直接或间接影响肾透明细胞癌中PTEN基因功能下调或失活的不同机制和影响因素,以期为临床治疗肾透明细胞癌提供新的理论依据。Renal clear cell carcinoma is the most common and deadly urological cancer in adult kidney cancer,in which abnormal activation of PI3K/Akt signaling pathway plays an important role in the occurrence and development of renal clear cell carcinoma.PTEN is a key inhibitory gene of PI3K/Akt signaling pathway,and abnormal activation of PI3K/Akt signaling pathway is caused by downregulation or inactivation of PTEN gene function.Therefore,understanding the mechanism and influencing factors of PTEN gene downregulation or inactivation in renal clear cell carcinoma is crucial for regulating abnormal activation of PI3K/Akt signaling pathway.This article aims to summarize different mechanisms and influencing factors that directly or indirectly affect downregulation or inactivation of PTEN gene function in renal clear cell carcinoma,in order to provide new theoretical basis for clinical treatment of renal clear cell carcinoma.
