桂西北壮族人群卵巢浆液性癌组织Twist和MGMT的表达及意义

摘　　要：目的研究Twist和MGMT在卵巢浆液性癌组织中的表达及意义,以便深入了解肿瘤发生发展的机制。方法选取300份活检组织样本,分别为良性卵巢囊肿(n=80)、交界性浆液性囊腺瘤(n=80)及卵巢浆液性癌(n=140)。采用免疫组织化学染色法检测Twist和MGMT蛋白表达情况,分析Twist、MGMT阳性和阴性患者临床病理因素的差异。结果在卵巢浆液性癌、交界性浆液性囊腺瘤和良性囊肿病变中,Twist蛋白的阳性率依次为76.43%、36.25%和28.75%,MGMT蛋白的阳性率分别为34.29%、48.75%和63.75%。卵巢浆液性癌患者中,Twist阳性患者淋巴结转移、分化程度低、Ⅲ+Ⅳ分期的占比高于阴性患者,MGMT阳性患者在淋巴结转移、Ⅲ+Ⅳ分期的占比均低于阴性患者(P<0.05)。结论在卵巢浆液性癌组织中,Twist表达上调,MGMT表达下调,Twist和MGMT的表达水平可能与肿瘤转移风险和肿瘤分期相关联。Objective To study the expression and significance of Twist and MGMT in ovarian serous carcinoma tissue,in order to further understand the mechanism of tumorigenesis and development.Methods 300 samples of benign ovarian cysts(n=80),borderline serous cystadenoma(n=80)and serous ovarian cancer(n=140)were selected.Immunohistochemical staining was used to detect the expression of Twist and MGMT protein,and the differences of clinicopathological factors between Twist and MGMT positive and negative patients were analyzed.Results In ovarian serous carcinoma,borderline serous cystadenoma and benign cyst,the positive rates of Twist protein were 76.43%,36.25% and 28.75%,and the positive rates of MGMT protein were 34.29%,48.75% and 63.75%,respectively.In patients with ovarian serous cancer,the proportion of lymph node metastasis,low differentiation and Ⅲ+Ⅳ stage in Twist positive patients were higher than those in negative patients,and the proportion of lymph node metastasis and Ⅲ+Ⅳ stage in MGMT positive patients were lower than those in negative patients(P<0.05).Conclusion In ovarian serous carcinoma tissue,the Twist expression is up-regulated and the MGMT expression is downregulated.The expression levels of Twist and MGMT may be related to the risk of tumor metastasis and tumor stage.
