摘 要:目的探讨肌肉生长抑制素(MSTN)对机械通气所致大鼠膈肌功能障碍的影响及可能机制。方法将20只SD大鼠随机平均分为正常对照组、机械通气24h组、机械通气48h组和机械通气72h组,每组5只。正常对照组自主呼吸,机械通气组根据分组予以不同时长的机械通气处理。观察各大鼠膈肌重量、血浆MSTN的表达水平,以及膈肌中MSTN、p-AKT/AKT和p-mTOR/mTOR等蛋白的表达情况。结果机械通气导致大鼠膈肌重量减轻,以及血浆中MSTN表达上调和p-AKT/AKT、p-mTOR/mTOR下调,变化呈时间依赖性(P<0.05)。结论MSTN在机械通气所致大鼠膈肌功能障碍中上调,可能与AKT/mTOR信号通路抑制相关。Objective To investigate the effect of MSTN on mechanical ventilation-induced diaphragmatic dysfunction in rats.Methods 20 SD rats were randomly divided into a normal control group,a 24-hour mechanical ventilation group,a 48-hour mechanical ventilation group and a 72-hour mechanical ventilation group,with 5 rats in each group.The normal control group had spontaneous breathing,while the mechanical ventilation group received mechanical ventilation treatment of different durations according to their grouping.The weight of the diaphragm in each rat was recorded,the expression level of plasma MSTN,and the expression of MSTN,p-AKT/AKT,and p-mTOR/mTOR proteins in the diaphragm were analyzed.Results It was found that in mechanical ventilation group there was a decrease of diaphragm weight in rats,as well as upregulation of MSTN expression and downregulation of p-AKT/AKT and p-mTOR/mTOR in plasma,with time-dependent changes(P<0.05).Conclusions MSTN was up-regulated in mechanical ventilation-induced diaphragmatic dysfunction in rats,which might be associated with inhibition of the AKT/mTOR signaling pathway.
