肺腺癌组织中miR-190-5p、PHLPP1的表达与病理参数和预后的关系

摘　　要：目的探讨肺腺癌(LUAD)组织中微小核糖核酸(miRNA)-190-5p、PH结构域和富亮氨酸的重复蛋白磷酸酶1(PHLPP1)的表达及与病理参数和预后的关系。方法选取上海健康医学院附属崇明医院收治的LUAD患者93例,根据miR-190-5p、PHLPP1 mRNA在LUAD的表达均值分为高、低表达组。采用实时荧光定量聚合酶链式反应检测LUAD组织与癌旁组织miR-190-5p、PHLPP1 mRNA表达。比较LUAD组织miR-190-5p、PHLPP1 mRNA表达在不同病理参数中差异。K-M法绘制不同miR-190-5p、PHLPP1 mRNA表达患者生存曲线。结果与癌旁组织比较,LUAD组织miR-190-5p表达升高,PHLPP1 mRNA表达降低(P<0.05)。PHLPP1与miR-190-5p在LUAD组织呈负相关(P<0.05)。LUAD组织miR-190-5p、PHLPP1 mRNA表达在不同分化程度、TNM分期、淋巴结转移中比较差异有统计学意义(P<0.05)。miR-190-5p高表达患者3年总生存率较低表达患者更低,PHLPP1 mRNA高表达患者3年总生存率较高于低表达患者更高(P<0.05)。结论LUAD组织miR-190-5p高表达和PHLPP1 mRNA低表达,与分化程度、TNM分期、淋巴结转移和不良预后有关。Objective To investigate the expression of microRNA-190-5p(miR-190-5p)and PH domain-containing leucine-rich repeat protein phosphatase 1(PHLPP1)in lung adenocarcinoma(LUAD)tissue and their correlation with pathological parameters and prognosis.Methods A total of 93 patients with LUAD who were treated at our hospital were selected and divided into high and low expression groups based on the mean levels of miR-190-5p and PHLPP1 mRNA in LUAD tissues.Real-time quantitative polymerase chain reaction was used to detect the expression of miR-190-5p and PHLPP1 mRNA in LUAD tissues and adjacent non-cancerous tissues.Differences in miR-190-5p and PHLPP1 mRNA expression between different pathological parameters in LUAD tissues were analyzed.The Kaplan-Meier(K-M)method was applied to plot survival curves for patients with different levels of miR-190-5p and PHLPP1 mRNA expression.Results Compared with adjacent tissues,miR-190-5p expression increased in LUAD tissues,while PHLPP1 mRNA expression decreased(P<0.05).A negatively correlated was observed between PHLPP1 and miR-190-5 in LUAD tissues(P<0.05).The expression of miR-190-5p and PHLPP1 mRNA in LUAD tissues differed significantly across levels of differentiation,TNM stage,and lymph node metastasis(P<0.05).Patients high miR-190-5p expression had a lower 3-year overall survival rate compared to those with low expression,while patients with high PHLPP1 mRNA expression showed a higher 3-year overall survival rate compared to those with low expression(P<0.05).Conclusion High expression of miR-190-5p and low expression of PHLPP1 mRNA in LUAD tissue are associated with differentiation,TNM stage,lymph node metastasis,and poor prognosis.
