摘  要:目的:以藏药诃子为研究对象,探究不同电子束辐照剂量对诃子主要活性成分和品质的影响。方法:开发以0.2%磷酸-甲醇(95∶5)为流动相,检测波长271 nm,柱温30℃,流速为1.0 mL/min的高效液相色谱法,用于检测诃子中没食子酸含量,并考察电子束辐照剂量对诃子活性成分、微生物含量及理化性质的影响。结果:没食子酸质量浓度在24.024~120.121μg/mL时,与峰面积线性关系良好(r=0.9999,n=5),精密度、重复性、稳定性和加标回收率试验结果的RSD均<2.0%(n=6);采用电子束辐照(0、3、5 kGy)处理诃子,辐照剂量为3 kGy及以上能显著降低微生物数量,并符合2020版《中华人民共和国药典》四部中非无菌产品微生物限度检查的规定,没食子酸和浸出物含量无显著差异(P>0.05),并且微生物数量、活性成分含量和浸出物含量在两个贮藏点间(0个月和10个月)无显著性差异。贮藏0个月,3、5 kGy处理组水分含量升高,贮藏10个月,辐照处理组水分降低,与CK无显著性差异。电子束辐照剂量≥3 kGy能达到良好的灭菌效果,保障诃子的药材品质。结论:电子束辐照可有效降低诃子药材中微生物的含量,同时能保持活性成分的稳定,电子束辐照可为诃子贮藏稳定性提供一定的理论参考。Objective:To investigate the effect of electron accelerator irradiation sterilization on Terminalia chebula Retz.,the effects of different electron beam irradiation doses(0,3,5 kGy)on the main active components and quality of Terminalia chebula Retz.were investigated.Methods:Determination by HPLC,the mobile phase was 0.2%phosphoric acid-methanol(95∶5)and the detection wavelength was 271 nm,the column temperature was 30℃,the flow rate was 1.0 mL/min.On this basis,the effects of electron beam irradiation dose on the active components,microbial content and physicochemical properties of Terminalia chebula Retz.were investigated.Results:The linear range of Terminalia chebula Retz.was 24.024~120.1212μg/mL(r=0.9999,n=5).The RSDs of precision,repeatability,stability and spiked recovery were less than 2.0%(n=6).Electron beam irradiation with doses of 0,3,5 kGy was used to treat Terminalia chebula Retz..The irradiation dose of More than 3 kGy significantly reduce the number of microorganisms and reach the microbial limit test of non-sterile products in the Chinese Pharmacopoeia(Edition 2020,VolumeⅣ).There was no significant difference in gallic acid and extract content(P>0.05),moreover there was no significant difference in the number of microorganisms,active ingredient content and extract content between the two storage points(0 month and 10 months).After 0 month of storage,the moisture content of the 3,5 kGy treatment group increased,and after 10 months of storage,the moisture content of the irradiation treatment group decreased,which was not significantly different from that of CK.The electron beam irradiation dose of 3 kGy can achieve sterilization effect and ensure the quality of Terminalia chebula Retz..Conclusion:This investigation is suitable for the sterilization of Terminalia chebula Retz.,which can provide a theoretical reference for the application of electron beam irradiation technology in Terminalia chebula Retz.storage.
