基于网络药理学与分子对接探讨小柴胡汤治疗感染后咳嗽的作用机制

摘　　要：目的:探究小柴胡汤治疗感染后咳嗽的中医理论与药理作用,为临床治疗感染后咳嗽提供参考依据。方法:筛选小柴胡汤中的中药活性成分的相关靶点,接着在疾病数据库中收集感染后咳嗽(PIC)的靶点基因,经拓扑分析,筛选小柴胡汤治疗PIC的关键成分,最后通过GO分析和KEGG分析明确小柴胡汤的治疗机制。结果:小柴胡汤与PIC存在200个相同靶点,经拓扑分析获得17个核心靶点,主要通过人巨细胞病毒感染、卡波西肉瘤相关疱疹病毒感染等通路治疗PIC,分子对接表明核心化合物与靶点具备良好的对接活性。结论:中医可从少阳论治感染后咳嗽,宜用小柴胡汤,其方中用药配伍既有升发疏散、祛邪之效,又有降气止咳化痰之能。基于网络药理学研究,小柴胡汤能够通过TP53、RelA、Stat3、AKT1、IL-6、MYC等靶点,调节感染后咳嗽疾病相关的蛋白酶活性、细胞衰老过程以及氧化应激等信号通路,进而发挥止咳、抗病毒等药效作用。Objective:To explore the traditional Chinese medicine theory and pharmacological effects of Xiao Chai Hu decoction in treating post-infection cough,so as to provide a reference for the clinical treatment of post-infection cough.Methods:Screened the active ingredient-related targets in Xiao Chai Hu decoction.Subsequently,target genes of the PIC were collected in the disease database.Through topological analysis,the key components of Xiaochaihu decoction in the treatment of PIC were screened.Finally,GO analysis and KEGG analysis were used to analyze the therapeutic mechanism of Xiao Chai Hu decoction.Results:Analysis indicates that Xiao Chai Hu decoction has a total of 200 same targets with PIC,and 17 core targets were obtained through topological analysis,mainly through human cytomegalovirus infection,Kaposi sarcoma-associated herpesvirus infection and other pathways for the treatment of PIC.Molecular docking showed that the core compound had good docking activity with the target.Conclusion:Traditional Chinese medicine can treat post-infection cough from the perspective of Shaoyang,using Xiao Chai Hu decoction.The ingredients in this formula are well-matched,with both the ability to ascend and disperse to expel pathogens,and the function to descend qi,relieve cough,and transform phlegm.Moreover,based on network pharmacology research,Xiao Chai Hu decoction can regulate the activity of proteases related to post-infection cough disease,cellular aging processes,and oxidative stress signaling pathways through targets such as TP53,RelA,Stat3,AKT1,IL-6,and MYC,thereby exerting therapeutic effects such as cough suppression and antiviral activities.
