游泳运动联合药物干预对木瓜蛋白酶诱导的膝关节骨性关节炎大鼠的影响

摘  要:目的:探讨游泳运动联合药物干预对木瓜蛋白酶诱导的膝关节骨性关节炎(KOA)大鼠血清转化生长因子-β1(TGF-β1)、基质金属蛋白酶-3(MMP-3)、血管内皮生长因子(VEGF)表达的影响。方法:SD大鼠50只,雄雌各半,分为正常对照组(正常组)、KOA模型对照组(模型组)、药物灌胃组(药物组)、游泳运动组(运动组)、游泳运动联合药物干预组(联合组),每组10只。KOA大鼠模型制备采用膝关节腔注射木瓜蛋白酶法,模型制备期4周。药物组和联合组灌胃盐酸氨基葡萄糖水溶液,剂量0.26g/kg,运动组和联合组进行游泳运动,50min/次,1次/d,6d/周,持续6周,末次实验结束24h后,股动脉取血处死大鼠,取膝关节进行病理学观察,ELISA法检测各组大鼠血清TNF-α、IL-1β、IL-6水平及TGF-β1、MMP-3、VEGF表达。结果:6周实验结束后,与正常组比较,模型组和各实验组大鼠血清TNF-α、IL-1β、IL-6水平及TGF-β1、MMP-3、VEGF表达显著升高(P<0.05),与模型组比较,运动组、药物组和联合组各项观测指标显著降低(P<0.05),相关性分析显示,KOA大鼠血清内TGF-β1、MMP-3、VEGF表达与TNF-α、IL-1β、IL-6水平呈显著正相关(P<0.05),病理观察显示,与模型组比较,运动组、药物组及联合组大鼠关节滑膜组织内炎细胞渗出、纤维组织增生等炎症病变程度均有所改善,其中联合组的炎症缓解程度更优于药物组。结论:游泳运动联合药物干预能够有效缓解木瓜蛋白酶诱导的KOA大鼠炎症状态,其作用机制可能与游泳运动进一步下调TGF-β1、MMP-3、VEGF表达有关。Objective:To explore the effect of swimming combined with drug intervention on serum in knee osteoarthritis(KOA)rats induced by papain effects of transforming growth factor-β1(TGF-β1),matrix metalloproteinase-3(MMP-3)and vascular endothelial growth factor(VEGF)expression.Methods:50 SD rats,with half male and half female,were divided into a normal control group(normal group),KOA model control group(model group),drug orally administered group(drug group),swimming exercise group(exercise group),and swimming exercise combined with drug intervention group(combination group),with 10 rats per group.The KOA rat model was prepared by injecting papain into the knee joint cavity,with a preparation period of 4 weeks.The drug group and combination group were orally administered with glucosamine hydrochloride aqueous solution at a dose of 0.26g/kg.The exercise group and combination group underwent swimming exercise at a rate of 50min/time,once a day,6 days a week,for a duration of 6 weeks.After 24 hours of the last experiment,blood was taken from the femoral artery to euthanize the rats,and the knee joints were taken for pathological observation.The expression of TGF-β1,MMP-3,VEGF and TNF-α,IL-1β,IL-6 levels in the serum of each group of rats was detected by ELISA.Results:After the completion of the 6 weeks experiment,compared with the normal group,the serum levels of the model group and each experimental group rats were measured TNF-α,IL-1β,IL-6 levels and the expression of TGF-β1,MMP-3 and VEGF was significantly increased(P<0.05),and compared with the model group,the observation indicators of the exercise group,drug group,and combination group were significantly reduced(P<0.05).Correlation analysis showed that the serum in KOA rats was significantly reduced TGF-β1,MMP-3,VEGF expression and the level of TNF-α,IL-1β,IL-6 showed a significant positive correlation(P<0.05),the pathological observations showed that compared with the model group,the exercise group,drug group,and combination group all improved the degree
