摘 要:以茶树菇CS-8、CS-15、CS-16为试材,采用继代培养方法,研究了不同继代次数对茶树菇菌丝的形态、生长速度、胞外酶活性、菌丝吃料快慢的影响,以期为后续茶树菇的生产提供参考依据。结果表明:随着继代次数的增加,茶树菇菌丝的生长速度逐渐降低、菌丝密度逐渐变稀疏、菌丝活力在F_(5)开始逐渐降低;淀粉酶活性随着继代次数的增加没有显著相关性;纤维素酶和蛋白酶在F_(1)就开始下降,木聚糖酶在F_(3)或F_(5)开始下降,漆酶在F_(5)或F_(7)开始下降;对各指标进行相关性分析,发现菌丝吃料快慢受母种菌丝生长速度、漆酶和纤维素酶活性影响,木聚糖酶与菌丝吃料速度呈负相关,蛋白酶和淀粉酶与茶树菇菌丝吃料无显著相关性或相关性不显著。因此,为确保菌种的优良性状及活性,继代次数应严格控制在F_(5)以内,且选择漆酶和纤维素酶活性高的菌株。Taking Agrocybe aegerita CS-8,CS-15and CS-16as the experimental materials,the effects of different subculture times on mycelia morphology,growth rate,extracellular enzyme activity and mycelia feeding speed of Agrocybe aegerita were studied,in order to provide reference for the subsequent production of Agrocybe aegerita.The results showed that with the increase of subgenerations,the growth rate of mycelia gradually decreased,the density of mycelia gradually became sparse,and the activity of mycelia gradually decreased in F_(5).Amylase activity had no significant correlation with the increase of subgenerations.Cellulase and protease began to decline in the F_(1),xylanase began to decline in F_(3)or F_(5),laccase began to decline in F_(5)or F_(7).Correlation analysis of each index showed that mycelia feeding speed was affected by parent mycelia growth rate,laccase and cellulase activities,xylanase was negatively correlated with mycelia feeding speed,protease and amylase were not significantly correlated with mycelia feeding.Therefore,in order to ensure the good properties and activities of the strains,the number of subgenerations should be strictly controlled within F_(5),and strains with high laccase and cellulase activities should be selected.
