摘　　要：目的分析瞬时感受器电位香草素受体4(transient receptor potential vanilloid 4,TRPV4)在胃癌中的表达及其临床意义。方法挖掘TCGA数据库中胃癌样本的临床数据、甲基化谱,观察其在胃癌中的表达水平及其甲基化程度,并用免疫组化观察胃癌患者组织样本中TRPV4的表达水平及其表达与临床病理特征的关系。用Kaplan-Meier plotter数据库探索TRPV4的表达与患者预后的关系,用TIMER数据库评估TRPV4与免疫细胞浸润的相关性。结果与非肿瘤组织比较,TRPV4在胃癌中高表达,高表达的TRPV4与胃癌患者的不良预后相关。TRPV4的表达与多个位点的甲基化密切相关。TRPV4的表达与CD8^(+)T细胞、NK细胞等的浸润相关。与TRPV4共表达的基因主要参与了免疫反应、抗原及脂肪酸的结合等生物学过程。结论TRPV4在胃癌中高表达,其表达量可能为影响胃癌预后不良的危险因素。Objective To investigate the expression of transient receptor potential vanilloid 4(TRPV4)in gastric cancer and its clinical significance.Methods Clinical data and methylation spectra of gastric cancer samples in TCGA database were excavated to observe the expression level and methylation level of TRPV4 protein in gastric cancer patients.The expression level of TRPV4 and its relationship with clinicopathological features in gastric cancer tissue samples were observed by immunohistochemistry.Kaplan-Meier plotter database was used to explore the relationship between TRPV4 expression and prognosis of patients,and TIMER database was used to evaluate the relationship between TRPV4 expression and immune cell infiltration.Results TRPV4 was highly expressed in gastric cancer compared with non-tumor tissues,and high expression of TRPV4 was associated with poor prognosis in gastric cancer patients.The expression of TRPV4 was closely related to methylation of multiple sites.The expression of TRPV4 was correlated with the infiltration of CD8^(+)T cells and NK cells.The genes co-expressed with TRPV4 were mainly involved in the biological processes such as immune response,antigen and fatty acid binding.Conclusion TRPV4 is highly expressed in gastric cancer,and its expression level may be a risk factor for poor prognosis of gastric cancer.
