摘  要:旨在分析短小芽孢杆菌SMU5927的基因组序列,并对该菌株的功能基因进行挖掘,为SMU5927的抑菌作用提供科学依据.通过提取菌株DNA,使用HiSeq平台建库及库检,Novaseq 6000测序平台对该菌株的测序分析,分别与GO、KEGG、COG和NR数据库进行基因功能注释.通过对测序结果的分析,SMU5927基因组总长度为5502707 bp,GC含量为40.98%,可编码的基因总长度为4788108 bp;对该菌株进行GO基因功能注释分析发现,其中5个基因参与免疫过程,2个基因与细胞增殖有关,11个基因与抗氧化活性有关;在KEGG数据库中共得到3440个基因的注释,6个基因与免疫相关,有5个基因与过氧化物酶体增殖激活受体(Peroxisome proliferator-activated receptors,PPAR)信号通路有关;COG分析发现有46个基因与主要协同转运蛋白超家族(Major facilitator superfamily,MFS)有关,从而保证了SMU5927的耐药性;通过NR分析,发现基因组中含有33种潜在的抑菌物质的相关基因.Surfactin、环状细菌素、Holin起到抗病原和抑菌的作用,MsrA、MsrB、PDF、NRPSs和MraY均有促进抗生素生成和制成抗菌剂的潜力;SppA、RF1、RF2和RF3能促进释放蛋白,SOD、CAT和SCO家族蛋白能防御氧化应激.综上所述,通过对SMU5927基因组序列功能注释的分析,从分子水平进一步验证了短小芽孢杆菌中存在抑菌物质,为该菌株的进一步开发利用提供了理论基础和科学依据.The aim was to analyze the genome sequence of Bacillus pumilus SMU5927 to excavate the functional genes of the strain.The DNA of the strain was extracted,and the library was established and checked on HiSeq platform.The strain was se-quenced and analyzed on Novaseq 6000 sequencing platform,and the gene function was annotated with GO,KEGG,COG and NR databases respectively.The total genome length of SMU5927 was 5502707 bp,the GC content was 40.98%,and the total length of the encoded gene was 4788108 bp.GO gene function annotation analysis of the strain showed that 5 genes were in-volved in immune process,2 genes were related to cell proliferation and 11 genes were related to antioxidant activity in SMU5927.There were 3440 genes annotated in KEGG database,and 6 genes were related to immunity,and there were 5 genes related to PPAR signaling pathway.Forty-six genes in COG were related to MFS transporter,which ensured the tolerance proper-ties of SMU5927.Through NR analysis,it was found that the genome contained 33 genes related to potential antibacterial sub-stances.Surfactin,cyclic bacteriocin and Holin played the role of pathogen resistance and bacteriostasis.MrsA,MsrB,PDF,NRPSs and MraY had the potential to promote the production of antibiotics and made antibacterial agents.SppA,RF1,RF2,and RF3 could promote the release of proteins.SOD,CAT,and SCO family proteins could defend against oxidative stress.In conclu-sion,through the analysis of the genome sequence of SMU5927,it was further verified that there were antibacterial substances in Bacillus pumilus at the molecular level.This study validated an important theoretical basis for the exploration of its function and antibacterial mechanism,which provided theoretical basis and scientific guidance for SMU5927 as a substitute for antibiotics.
