摘  要:叶片的叶绿素含量与农作物的光合作用效率和产量潜力紧密相关。为探究影响甘蓝型油菜叶绿素含量的遗传机理,挖掘与甘蓝型油菜苗期叶绿素含量显著相关的单核苷酸多态性(SNP)位点及候选基因,本研究对278份来自世界各地的甘蓝型油菜核心种质苗期子叶中叶绿素的含量进行了测定,并利用rMVP软件的GLM、MLM和FarmCPU三种全基因组关联分析模型对总叶绿素含量、叶绿素a含量、叶绿素b含量及叶绿素a与叶绿素b含量比值分别进行了全基因组关联分析。研究结果显示:叶绿素a与叶绿素b含量比值的变异系数最小,为20%,叶绿素b含量的变异系数最大,达到了33%;上述四个叶绿素含量相关表型分别检测到了2、6、4和37个显著关联的SNP位点,这些SNP位点集中分布在A03、A04、C03、C05和C07等染色体上。进一步结合基因注释和同源比对共鉴定到70个候选基因,全生育期表达谱分析发现其中12个候选基因在叶片和角果等绿色组织中特异性高表达。实时荧光定量PCR(qRTPCR)结果显示,其中9个候选基因的表达量在高或低叶绿素含量品种中存在差异,暗示这些候选基因可能参与了叶绿素含量的调控。本研究结果有利于进一步揭示叶绿素含量调控的遗传基础,为甘蓝型油菜高光效育种提供了理论基础和种质资源。The chlorophyll content of leaves is closely related to the photosynthesis efficiency and the yield potential of crops.To explore the genetic mechanism affecting the chlorophyll content of Brassica napus and to identify SNP loci and candidate genes significantly related to the chlorophyll content at the seedling stage,the chlorophyll contents at the seedling stage of 278 core germplasms of Brassica napus from all over the world were measured.GLM,MLM,and FarmCPU association models in rMVP software were employed to conduct the genome-wide association study of total chlorophyll content,chlorophyll a content,chlorophyll b content,and the ratio of chlorophyll a to chlorophyll b content.The results showed that the rapeseed population exhibited a significant degree of diversity in chlorophyll content,with the highest coefficient of variation for chlorophyll b content at 33%and the ratio of chlorophyll a to chlorophyll b having the lowest coefficient of variation at 20%.There were 2,6,4,and 37 SNPs were detected to be significantly associated with four chlorophyll-content-related traits,respectively.These associated SNPs were clustered on chromosomes A03,A04,C03,C05,and C07.A total of 70 candidate genes were identified through gene annotation and homology comparison analyses.Whole growth period expression profiling analysis revealed that 12 candidate genes were preferentially expressed in green tissues,including leaves and siliques.The results of qRT-PCR revealed that the expression levels of nine candidate genes varied significant between high and low chlorophyll content varieties,suggesting that these genes may be involved in the regulation of chlorophyll content.This study will help to uncover the genetic basis of chlorophyll content regulation and provide a theoretical basis and germplasm resources for high photosynthetic efficiency breeding of Brassica napus.
