摘  要:为探究大型海藻中重金属的富集效应和重金属离子间的相互作用及其对藻体内金属结合蛋白的诱导影响,本研究以羊栖菜幼苗为研究对象,分别采用单一重金属(Cr、Pb、Cu、Cd)和复合重金属体系在低浓度和高浓度下进行室内胁迫培养,研究不同重金属离子在藻体中的富集能力,以及金属硫蛋白(MTs)和谷胱甘肽(GSH)的表达趋势。结果表明,羊栖菜藻体内Cr、Pb、Cu、Cd的富集量与胁迫浓度和富集时间相关,胁迫浓度越高、富集时间越长,富集量越高;不同重金属的富集能力存在差异,Pb的富集能力最强、Cr和Cu次之、Cd最弱;同种金属在低浓度下的富集能力大于高浓度下的富集能力。此外,在富集过程中发现低浓度下Cu和Cd共存表现为拮抗作用,Pb和Cu、Pb和Cd之间表现为协同作用,而高浓度下Pb和Cd呈拮抗作用、Pb和Cu之间的协同作用减弱、Cu和Cd之间仍呈拮抗作用。试验周期内,受低浓度单一金属和二元复合金属元素诱导后,藻体内的MTs和GSH含量随诱导时间延长呈先升后降趋势;四元复合金属体系诱导后,MTs和GSH含量显著低于一元和二元金属诱导。高浓度单一和复合金属诱导均会显著抑制MTs和GSH的诱导表达。本研究可为大型经济海藻的规模养殖及水体重金属污染监测和控制等提供理论基础和科学依据。In order to study the accumulation and interaction of heavy metals in macroalgae,as well as their induced effects on metal-binding proteins,with seedlings of Sargassum fusiforme indoor cultivation,the accumulation ability of different heavy metals as well as the expression trends and differences of metallothionein(MTs)and glutathione(GSH)were studied under single heavy metals(Cr,Pb,Cu,Cd)and mixed heavy metals at lower and higher concentrations.The results showed that the accumulation contents of Cr,Pb,Cu and Cd in Sargassum fusiforme were related to the induced heavy metal concentrations,accumulation time and coexisting ions.The accumulation contents of heavy metals in Sargassum fusiforme were higher with higher treatment concentrations and longer accumulation time.There were differences in the accumulation ability among different heavy metals,which of Pb had the strongest,followed by Cr and Cu,and Cd was the weakest.The accumulation ability of the same metal at lower concentrations was greater than that at higher concentrations.In addition,the Cu had an antagonistic effect when coexisted with Cd at lower concentrations,while there was a synergistic effect between Pb and Cu,as well as Pb and Cd.At higher concentrations,Pb and Cd represented an antagonistic effect,and the synergistic effect between Pb and Cu weakened,while Cu and Cd still represented an antagonistic effect.During the experimental periods,MTs and GSH contents induced by single heavy metal and binary mixtures both showed a trend of first increasing and then decreasing at lower concentrations with increasing induced time,while the contents of MTs and GSH were significantly decreased after the quaternary mixtures treatments compared with the treatments of single heavy metal and binary mixtures.Higher concentrations of both single and mixed heavy metals significantly inhibited the induced expression of MTs and GSH.This study provides theoretical and scientific bases for large-scale economic seaweed cultivation,as well as controlling and evaluating hea
