摘　　要：目的构建一个基于中性粒细胞胞外陷阱相关基因的预测胶质瘤患者预后的列线图模型。方法本研究纳入来自中国脑胶质瘤基因组图谱(CGGA)1018例患者(训练组693例,验证组325例)的临床病理信息和转录组测序数据。应用LASSO-COX算法筛选出与胶质瘤患者总生存期相关的基因并建立预后风险评分方程。使用校准曲线和C指数来评估列线图的性能。结果基于单因素和多因素Cox分析得到了10个中性粒细胞胞外陷阱相关基因。列线图结合了基因信息和临床病理预测因子,在训练组和验证组中显示出良好的预后能力。此外,本研究还发现复发性胶质瘤中肿瘤细胞富集了一种能够正向调控白细胞介素功能的机制。结论本研究开发的列线图模型可以预测胶质瘤患者的1、2、3、5和10年生存率。Objective To construct a nomogram model for predicting the prognosis of glioma patients based on neutrophil extracellular trap-related genes.Methods This study included clinicopathological information and transcriptome sequencing data from 1018 patients(693 patients in the training group and 325 patients in the validation group)from the Chinese Glioma Genome Atlas(CGGA).The LASSO-COX algorithm was used to screen out genes related to the overall survival of glioma patients and establish a prognostic risk scoring equation.Calibration curves and C-indexes were used to evaluate the performance of the nomogram.Results Based on univariate and multivariate Cox analysis,10 neutrophil extracellular trap-related genes were obtained.The nomogram combined genetic information and clinicopathological predictors,showing good prognostic ability in the training group and the validation group.In addition,this study also found that tumor cells in recurrent glioma enriched a mechanism that could positively regulate interleukin function.Conclusion The nomogram model developed in this study can predict the 1-year,2-year,3-year,5-year,and 10-year survival rates of glioma patients.
