摘 要:目的分析人同源盒基因D8(HOXD8)在脑胶质瘤中的表达特征,探讨其与肿瘤进展、患者生存预后以及免疫微环境的关系。方法整合TCGA数据库中的胶质瘤样本数据,进行生物信息学分析,研究HOXD8的表达模式,通过实时荧光定量PCR验证其在胶质瘤组织中的表达差异;结合单基因生存分析、Kaplan-Meier生存曲线及免疫细胞浸润分析,探讨HOXD8表达对患者预后和肿瘤免疫微环境的影响;运用KEGG通路富集分析和GO基因功能注释分析评估HOXD8的潜在分子机制。结果(1)HOXD8在胶质瘤组织中显著高表达,且表达水平随肿瘤分期的进展逐渐升高(均P<0.001)。(2)高表达HOXD8与患者总体生存率呈负相关(P<0.001)。(3)免疫细胞浸润分析显示,HOXD8高表达的肿瘤组织中,巨噬细胞及未成熟的树突状细胞等免疫抑制细胞浸润显著增加(均P<0.001)。(4)KEGG通路分析显示,HOXD8相关基因主要富集于细胞周期调控、DNA复制等信号通路。(5)GO功能分析揭示,HOXD8与DNA转录调控、核分裂和染色体分离等重要生物学过程密切相关。结论HOXD8在脑胶质瘤中显著高表达,且其表达与肿瘤分期、免疫细胞浸润水平及患者生存预后紧密相关,HOXD8可能通过调控肿瘤免疫微环境和细胞周期进程,促进胶质瘤的恶性进展。Objective This study aims to analyze the expression characteristics of the human homeobox gene D8(HOXD8)in gliomas and explore its relationship with tumor progression,patient survival prognosis,and the tumor immune microenvironment.Methods Glioma sample data from the TCGA database were integrated for bioinformatics analysis to study the expression pattern of HOXD8.Real-time quantitative PCR was used to verify the expression differences of HOXD8 in glioma tissues.Furthermore,single-gene survival analysis,Kaplan-Meier survival curves,and immune cell infiltration analysis were performed to systematically investigate the impact of HOXD8 expression on patient prognosis and the tumor immune microenvironment.Additionally,KEGG pathway enrichment analysis and GO gene function annotation were utilized to assess the potential molecular mechanisms of HOXD8.Results(1)HOXD8 was significantly overexpressed in glioma tissues,and its expression levels increased with tumor staging progression(all P<0.001).(2)High HOXD8 expression was significantly associated with poor overall survival in patients(P<0.001).(3)Immune cell infiltration analysis revealed that tumors with high HOXD8 expression exhibited significantly increased infiltration of immunosuppressive cells,such as macrophages and immature dendritic cells(all P<0.001).(4)KEGG pathway analysis showed that HOXD8-related genes were mainly enriched in cell cycle regulation and DNA replication pathways.(5)GO analysis revealed that HOXD8 is closely associated with DNA transcription regulation,nuclear division,and chromosome segregation.Conclusion HOXD8 is significantly overexpressed in gliomas,and its expression is closely related to tumor staging,immune cell infiltration levels,and patient survival prognosis.HOXD8 may promote glioma progression by regulating the tumor immune microenvironment and cell cycle processes,making it a potential target for prognosis evaluation and therapeutic intervention in gliomas.
