摘  要:目的:建立一种准确、快速、高效地测定海南番石榴中21种游离氨基酸含量的超高效液相色谱—串联质谱法。方法:番石榴样品经充分匀浆后用水超声提取,50%乙腈水溶液稀释后经ACQUITY UPLC BEH C_(18)(1.7μm,2.1 mm×100 mm)色谱柱分离,乙腈-0.2%甲酸水溶液体系等度洗脱,采用电喷雾离子源正离子模式扫描,多反应监测模式进行检测。结果:21种游离氨基酸在各自相应线性范围内,呈良好线性关系,相关系数(r2)均大于0.992,3个浓度水平的加标回收率为72%~116%,相对标准偏差为1.9%~7.2%,方法检出限为0.3~7.8μg/kg,方法定量限为1~25μg/kg。结论:该方法操作简单、灵敏度高、准确稳定,适用于海南番石榴中游离氨基酸的检测。【Objective】To establish an accurate,rapid and efficient ultra performance liquid chromatography-tandem mass spectrometry method for the determination of 21 kinds of free amino acids in guava of Hainan.【Method】The guava samples were fully homogenized and extracted with ultrasonic water.The samples were diluted with 50%acetonitrile solution by ACQUITY UPLC BEH C_(18)(1.7μm,2.1 mm×100 mm)column separation,acetonitrile-0.2%formic acid solution system iso-degree elution,electrospray ion source positive ion mode scanning,multi-reaction monitoring mode detection.【Result】The 21 free amino acids showed a good linear relationship in their respective linear ranges,and the correlation coefficients(r2)were all greater than 0.992.The recoveries of three concentrations ranged from 72%to 116%,the relative standard deviations were 1.9%to 7.2%,the detection limits were 0.3 to 7.8μg/kg,and the limits of quantitation were 1 to 25μg/kg.【Conclusion】The method is simple,sensitive,accurate and stable,which is suitable for the detection of free amino acids in guava.
