藏羚羊源丁酸梭菌的分离鉴定及全基因组分析

摘  要:采用厌氧微生物分离培养技术从藏羚羊粪便中分离出多株细菌,通过形态学观察、生理生化试验和16S rRNA测序技术,鉴定出1株丁酸梭菌FC36。为探究分离菌株潜在的益生功能,对其全基因组序列进行测定。同时采用体外试验对其益生功能进行研究,包括生长曲线与pH变化曲线测定、产淀粉酶与短链脂肪酸检测、耐受性能评估、抑菌试验。结果表明:该基因组大小4.4 Mbp,G+C含量28.53%,包含4022个蛋白质编码基因,共注释到532个碳水化合物酶类家族基因、1070个新陈代谢类通路相关基因、3244个同源蛋白簇基因及2个细菌素编码基因。菌株FC36生长速度较快,可产生淀粉酶和多种短链脂肪酸,耐受能力强,对大肠杆菌和沙门菌具有一定的抑制能力。综上,分离得到1株性能优良的丁酸梭菌,可为临床相关益生菌制剂的研发奠定基础。In this study,multiple bacterial strains were isolated from the feces of Pantholops hodgsoni by anaerobic microbial isolation and culture technology.A strain of Clostridium butyricum FC36 was identified by morphological observation,physiological and biochemical tests,and 16S rRNA sequencing technology.In order to explore the potential probiotic function of isolated strain,its whole genome sequence was determined.Assembly and functional analysis showed that the genome size was 4.4 Mbp,G+C content was 28.53%,and contained 4022 protein-coding genes.A total of 532 carbohydrate-enzyme family genes,1070 metabolic pathway related genes,3244 homologous protein cluster genes,and 2 bacteriocin coding genes were annotated.At the same time,in vitro experiments were carried out to study its probiotic function,including growth curve and pH change curve measurement,amylase production and short-chain fatty acid detection,tolerance evaluation of bacteriostatic test.The results showed that Clostridium butyricum FC36 had a fast growth rate,could produce amylase and a variety of short-chain fatty acids,had strong tolerance,and had certain inhibitory ability against Escherichia coli and Salmonella.In summary,a strain of Clostridium butyricum with excellent performance was isolated in this study,which laid a foundation for the development of relevant probiotics in clinical practice.
