辽宁省沈阳市猫细小病毒VP2基因克隆与遗传进化分析

摘　　要：为了解沈阳市猫细小病毒(FPV)的遗传进化情况,探究FPV免疫失败的原因,收集2022年该地区FPV感染猫肛拭子,采用PCR扩增FPV VP2基因,对VP2基因测序并进行同源性比对、主要编码氨基酸位点分析与遗传进化树绘制。结果显示;获得了7个FPV VP2基因片段,全长均为1755 bp,与50株参考毒株VP2基因同源性为98.8%~99.9%;与标准株和疫苗株相比,6个获得基因编码的VP2蛋白第91位点发生了改变,其中2个获得基因编码的VP2蛋白第295位点发生了改变;遗传进化树显示,FPV VP2基因分为3个基因群,7个获得基因中有6个位于G1群、1个位于G3群,与国内近年来流行毒株亲缘关系近,但与疫苗株和标准株均不在同一个基因群(G2群)。结果说明;沈阳市流行的FPV逐渐进化形成了不同于疫苗株和标准株的基因群,因此有必要加强FPV感染的流行病学监测,研发针对性更强的疫苗。In order to investigate genetic evolution of feline parvovirus(FPV)in Shenyang City,and to find out the reasons for the failure of immunization against FPV,anal swab samples were collected from infected cats in the city in 2022 to amplify FPV VP2 gene using PCR,followed by gene sequencing,homologous comparison,analysis on main amino acid sites and phylogenetic tree mapping.The results showed that 7 FPV VP2 gene fragments were obtained with the same full length of 1755 bp,and 98.8%-99.9%identical to sequences of VP2 genes of 50 reference strains;compared with standard and vaccine strains,the 91st amino acid sites of 6 VP2 proteins were with mutations,and the 295th amino acid sites in 2 of 6 VP2 proteins also changed.Phylogenetic tree showed that FPV VP2 gene was divided into 3 gene groups,including 6 obtained genes in this study located in G1 group,and 1 in G3 group,closely relating to current strains prevalent in China,but was not in the same gene group(G2 group)as the standard and vaccine strains.In conclusion,the FPV prevalent in Shenyang gradually evolved into the gene group different from the standard and vaccine strains,requiring to increase epidemiological surveillance over FPV infection,and to develop vaccines with more pertinence.
