摘  要:目的:研究丹皮酚对乙醇诱导的神经元损伤的影响。方法:取出生24 h内的SD乳鼠,分离培养皮质原代神经元。取第10天神经元,分为空白对照组、乙醇组和乙醇+丹皮酚组,分别用含PBS、1.5 g/L乙醇和1.5 g/L乙醇+100μmol/L丹皮酚溶液的培养基培养12 h。JC-1探针法检测线粒体膜电位,CCK-8法检测神经元活力,Annexin V/PI双染法检测神经元凋亡情况,WST-1法测定细胞培养液中超氧化物歧化酶(SOD)活性,TBA法测定细胞培养液中丙二醛(MDA)含量,Western blot法检测Bcl-2、Bax和cleaved-Caspase 3蛋白的表达。结果:与空白对照组相比,乙醇组线粒体膜电位降低,神经元活力和SOD活性降低,MDA含量增加,凋亡神经元增多,Bcl-2蛋白表达降低,Bax和cleaved-Caspase 3蛋白表达升高(P<0.05)。与乙醇组相比,乙醇+丹皮酚组线粒体膜电位升高,神经元活力和SOD活性增加,MDA含量降低,凋亡神经元减少,Bcl-2蛋白表达增加,Bax和cleaved-Caspase 3蛋白表达降低(P<0.05)。结论:丹皮酚可能通过抑制氧化应激和拮抗线粒体凋亡,缓解乙醇诱导的神经元损害。Aim:To explore the effects of paeonol on ethanol-induced neurons injury.Methods:Primary cortical neurons were isolated from SD neonatal rats within 24 hours of birth.The neurons cultured for 10 days were allocated into blank control group,ethanol group,and ethanol+paeonol group,and were cultured for 12 h with PBS,1.5 g/L ethanol,and 1.5 g/L ethanol+100μmol/L paeonol,respectively.Then,mitochondrial membrane potential was detected by JC-1 probe method;cell viability was detected by CCK-8 method;cell apoptosis was detected by Annexin V/PI staining;superoxide dismutase(SOD)activity and malondialdehyde(MDA)content in the culture supernatant were determined by WST-1 method and TBA method,respectively;the expression of Bcl-2,Bax and cleaved-Caspase 3 proteins was detected by Western blot.Results:Compared with the blank control group,the neuron viability and SOD activity of the ethanol group decreased,MDA content increased,the mitochondrial membrane potential decreased,the proportion of apoptosis neurons increased,the expression of Bcl-2 decreased,and the expression of Bax and cleaved-Caspase 3 increased(P<0.05).Compared with those of the ethanol group,the neuron viability and SOD activity of the ethanol+paeonol group increased,MDA content decreased,the mitochondrial membrane potential increased,the proportion of apoptosis neurons decreased,the expression of Bcl-2 increased,and the expression of Bax and cleaved-Caspase 3 decreased(P<0.05).Conclusion:Paeonol may alleviate ethanol-induced neurons damage by inhibiting oxidative stress and antagonizing mitochondrial apoptosis.
