摘  要:目的:探讨生长抑素联合大黄调控磷酸酶Ⅰ(Reg-1)促进急性胰腺炎(AP)大鼠胰岛β细胞再生的相关机制,为AP提供中西医干预靶点和理论依据。方法:健康清洁级8~10月龄SD雄性大鼠60只随机分为6组,即空白对照组、模型组、生长抑素组、大黄组、联合组与联合+Reg-1抑制组,每组10只。空白对照组不做任何处理,正常饲养;其他5组均采用5%牛磺胆酸钠溶液逆行注入胰胆管复制AP模型;模型组注射等量生理盐水,生长抑素组采用股静脉恒量泵1μg/kg注入生长抑素,大黄组采用10%大黄(1ml/200 g)灌胃,联合组采用生长抑素+大黄剂量同上,联合+Reg-1抑制组采用siRNA技术构建Reg-1抑制剂,经腹腔注射(1ml/200 g)。各组持续干预7 d,每天1次,记录大鼠的精神、活动以及存活情况。结果:各组大鼠均存活至实验结束。酶联免疫吸附试验(ELISA)检测静脉血炎性因子比较发现,各组实验前白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平差异无统计学意义(P>0.05);实验结束后空白对照组IL-6和TNF-α显著低于其他各组,模型组最高,联合组低于生长抑素组、大黄组和联合+Reg-1抑制组,差异均有统计学意义(P<0.05)。实验结束后空白对照组血淀粉酶、白细胞计数和中性粒细胞百分比显著低于其他各组,模型组最高,联合组低于生长抑素组、大黄组和联合+Reg-1抑制组,差异均有统计学意义(P<0.05)。实验结束后处死大鼠取得新鲜胰腺组织制作标本,采用流式细胞术检测β细胞数量比较发现,空白对照组凋亡率显著低于其他各组,模型组最高,联合组低于生长抑素组、大黄组和联合+Reg-1抑制组,差异均有统计学意义(P<0.05)。Western印迹检测胰腺组织凋亡蛋白Bax、Bcl-2和Caspase-3相对表达量比较发现,空白对照组显著低于其他各组,模型组最高,联合组低于生长抑素组、大黄组和联合+Reg-1抑制组,差异均有统计学意义(P<0.05)。Western印迹�Objective To investigate the mechanism of somatostatin combined with chinese rhubarb on the promotion of pancreatic isletsβcell regeneration in rats with acute pancreatitis(AP)by regulating phosphataseⅠ(Reg-1),so as to provide the target and theoretical basis for the intervention of traditional Chinese and western medicine for AP.Method 60 healthy male SD rats aged 8~10 months were randomly divided into 6 groups,namely blank control group,model group,somatostatin group,rhubarb group,combination group and combination+Reg-1 inhibition group,with 10 rats in each group.The blank control group was fed normally without any treatment;in the other 5 groups,5%sodium taurocholate solution was retrograde injected into the pancreaticobiliary duct to replicate AP model;the model group was injected with the same amount of normal saline,and the somatostatin group was injected with 1μg/kg of somatostatin with femoral vein constant volume pump,rhubarb group was given 10%rhubarb(1 ml/200 g)by gavage,combination group was given somatostatin+rhubarb,the dose was the same as above,and combination+Reg-1 inhibition group was injected intraperitoneally(1 ml/200 g)Reg-1 inhibitor constructed by siRNA technology.Each group continued to intervene for 7 days,once a day,and recorded the spirit,activity and survival of rats.Results All rats survived until the end of experiment.The inflammatory factors in venous blood detected by ELISA interleukin 6(IL-6)and tumor necrosis factor-α(TNF-α)before experiment in each group showed that there were no differences among the six groups(P>0.05);they were in blank control group after experiment significantly lower than other groups,the highest in model group,and lower in combination group than somatostatin group,rhubarb group and combination+Reg-1 inhibition group(P<0.05).The blood amylase,white blood cell count and neutrophil percentage in blank control group were significantly lower than those in other groups,the model group were the highest,while the combination group was lower than somatostatin
