摘 要:本研究开发了一种柱前自动化衍生-高效液相色谱-紫外法(柱前自动化衍生HPLC-UV)用于同时检测医疗美容产品中18种氨基酸。该方法以AdvanceBio AAA为分离色谱柱,磷酸氢二钠-四硼酸钠缓冲液和有机相[V(乙腈)∶V(甲醇)∶V(水)=45∶45∶10]为流动相进行梯度洗脱,在262、338 nm双波长下成功实现18种氨基酸的同时检测。研究结果表明,18种氨基酸在10~1000 nmol/mL浓度范围内,峰面积与浓度之间的线性关系良好(r≥0.99),检出限(LOD)和定量限(LQD)分别在0.2~9.6 nmol/mL和0.8~22.5 nmol/mL。在胶原蛋白和透明质酸酸钠复合溶液产品的标示含量范围内进行5个浓度水平的加标回收率(n=15)考察,18种氨基酸的平均回收率为92.3%~103.4%,RSD为0.1%~5.8%。此外,应用本方法对注射类医疗美容产品进行实际样品检测,结果与经验证的LC-MS/MS所得结果一致。本方法具有低成本、高灵敏、强专属性、定量结果准确和操作简单的特点,适用于常见注射用医疗美容产品中氨基酸的定量检测,可为生产企业或相关机构进行日常监测提供有力的技术支持。This study developed a pre column automated derivatization high performance liquid chromatography ultraviolet method(pre column automated derivatization HPLC-UV)for simultaneous detection of 18 amino acids in medical beauty products.This method uses AdvanceBio AAA as the separation chromatography column,disodium hydrogen phosphate sodium tetraborate buffer,and organic phase[V(acetonitrile):V(methanol):V(water)=45:45:10]as the mobile phase for gradient elution,and successfully achieves simultaneous detection of 18 amino acids at dual wavelengths of 262 and 338 nm.The research results showed that there was a good linear relationship between peak area and concentration(r≥0.99)for 18 amino acids in the concentration range of 10~1000 nmol/mL.The detection limit(LOD)and quantification limit(LQD)were 0.2~9.6 nmol/mL and 0.8~22.5 nmol/mL,respectively.Within the labeled content range of collagen and sodium hyaluronate composite solution products,five concentration levels of spiked recovery rates(n=15)were investigated.The average recovery rate of 18 amino acids was 92.3%~103.4%,with RSD ranging from 0.1%~5.8%.In addition,this method was applied to actual sample testing of injectable medical beauty products,and the results were consistent with the validated LC-MS/MS.This method has the characteristics of low cost,high sensitivity,strong specificity,accurate quantitative results,and simple operation.It is suitable for the quantitative detection of amino acids in common injectable medical beauty products and can provide strong technical support for daily monitoring by production enterprises or related institutions.
