摘 要:目的应用Box-Behnken响应面法设计,结合层次分析-熵权法优化草珊瑚的杀青工艺。方法以新绿原酸、刺五加苷B1、异嗪皮啶、落新妇苷和迷迭香酸的含量及总黄酮、总酚酸得率为评价指标,采用层次分析-熵权法确定各评价指标的综合权重系数进行评价分析,优选杀青工艺;以杀青温度、杀青次数、杀青时间为关键工艺参数,采用单因素试验确定各因素水平;Box-Behnken响应面法设计构建三因素三水平试验设计方案,结合综合权重系数进行综合评价,并通过DPPH自由基清除试验对杀青草珊瑚抗氧化的能力进行评价,通过设计空间思想优化工艺水平。结果草珊瑚最佳的杀青方法是热风杀青,响应面优化得到最佳工艺条件为杀青温度60℃,杀青次数2次,杀青时间50s。DPPH自由基清除试验结果表明,草珊瑚经过热风杀青后可以提升其体外抗氧化活性。建立设计空间为次数1~2次,温度60~62℃,时间55~65s。结论优化的草珊瑚热风杀青工艺合理、稳定、可行,可为草珊瑚的初加工提供理论与试验依据。OBJECTIVE To optimize the heat-process technology of Sarcandra glabra by combination of Box-Behnken design-response surface method and AHP-entropy weight method.METHODS The contents of neochlorogenic acid,eleutheroside B1,isofraxidin,astilbin and rosmarinic acid,as well as the yield of total flavonoids and total phenolic acids were used as evaluation indexes.The AHP-entropy weight method was used to determine the comprehensive weight coefficient of each evaluation index for evaluation and analysis.And the heat-process technology was optimized.With temperature,frequency and time as the key process parameters,the level of each factor was determined by single factor experiment.The Box-Behnken designresponse surface method was used to construct the three-factor and three-level experimental design scheme,and the comprehensive evaluation was carried out by combining the comprehensive weight coefficient.The DPPH free radical scavenging experiment was conducted to verify the effect of green removal on the antioxidant capacity of grass coral.Established a design space for process validation.RESULTS The optimal processing method for Sarcandra glabra was hot air drying process.The optimal process conditions obtained through response surface optimization were a blanching temperature of 60℃,a blanching frequency of 2 times,and a blanching time of 50 s.DPPH free radical scavenging experiment verified that grass coral could enhance its antioxidant activity in vitro after hot processing.The design space was established as follows:1-2 times,60-62℃,55-65 s.CONCLUSION The optimized heat-process technology is reasonable,stable and feasible,which can provide theoretical and experimental basis for the initial processing of Sarcandra glabra.
