摘  要:目的探讨双硫仑通过调控miR-767-5p表达对乳腺癌细胞的增殖、迁移和侵袭的影响及作用机制。方法不同浓度的双硫仑处理乳腺癌细胞MCF-7后,应用细胞计数试剂盒(CCK-8)检测细胞活力,Transwell实验检测细胞迁移和侵袭,实时荧光定量PCR(RT-qPCR)检测miR-767-5p表达水平。将miR-767-5p抑制物转染MCF-7细胞,检测下调miR-767-5p对双硫仑处理的MCF-7细胞增殖、迁移和侵袭的影响。结果双硫仑处理后MCF-7细胞的增殖、迁移和侵袭能力显著降低,miR-767-5p表达显著下降,并呈显著的量效关系(P<0.05)。下调miR-767-5p后MCF-7细胞的增殖、迁移和侵袭能力显著降低(P<0.05)。结论双硫仑对乳腺癌细胞的增殖、迁移和侵袭有抑制作用,其机制可能与下调miR-767-5p表达有关。Objective To investigate the effects of disulfiram on the proliferation,migration,and invasion of breast cancer cells by regulating the expression of miR-767-5p and its mechanism.Methods After MCF-7 cells were treated with different concentrations of disulfide,cell viability was detected by CCK-8,cell migration and invasion were detected by Transwell assay,and miR-767-5p expression levels were detected by real-time fluorescent quantitative PCR(RT-qPCR).miR-767-5p inhibitor was transfected into MCF-7 cells,and the effects of down-regulation of miR-767-5p on proliferation,migration,and invasion of disulfiram-treated MCF-7 cells were detected.Results The proliferation,migration,and invasion ability of MCF-7 cells were significantly decreased after treated by disulfiram,and the expression of miR-767-5p was significantly decreased,with a significant dose-effect relationship(P<0.05).The proliferation,migration,and invasion ability of MCF-7 cells were significantly decreased after miR-767-5p down-regulation(P<0.05).Conclusion Disulfiram can inhibit the proliferation,migration,and invasion of breast cancer cells,and its mechanism may be related to the down-regulation of miR-767-5p expression.
