摘 要:目的:口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)是口腔最常见的恶性肿瘤,术后易复发且预后较差。微RNA-320a(microRNA-320a,miR-320a)及SRY相关高迁移率族蛋白4(SRY-like high-mobility group box 4,SOX4)与肿瘤的发生、发展及预后密切相关。本研究旨在分析miR-320a和SOX4在OSCC组织中的表达及其与预后的相关性。方法:分别收集62例OSCC患者的肿瘤组织(OSCC组)、30例白斑、红斑或扁平苔藓患者的颊部病变组织(癌前病变组)和8例健康志愿者的正常口腔黏膜组织(对照组),并检测各组miR-320a和SOX4的表达水平,分析二者的相关性,及其与OSCC患者临床病理特征和术后总生存期(overall survival,OS)的相关性。结果:与对照组比较,癌前病变组和OSCC组的miR-320a表达水平均降低(均P<0.05),SOX4蛋白表达水平均升高(均P<0.05);与癌前病变组比较,OSCC组的miR-320a表达水平降低(P<0.05),SOX4蛋白的表达水平升高(P<0.05)。OSCC患者的miR-320a水平与SOX4水平呈负相关(r=−0.282,P<0.05)。MiR-320a和SOX4水平与OSCC患者的肿瘤淋巴结转移(tumor node metastasis,TNM)分期均有关(均P<0.05),且高miR-320a患者的OS高于低miR-320a患者(P<0.05),而高SOX4患者的OS低于低SOX4患者(P<0.05);miR-320a高表达是OSCC患者OS的保护因素(P<0.05)。结论:MiR-320a及SOX4的异常表达可能参与OSCC的发生、发展,miR-320a可作为OSCC患者的潜在预后指标。Objective:Oral squamous cell carcinoma(OSCC)is the most common malignant tumor in the oral cavity,characterized by a high recurrence rate and poor prognosis.MicroRNA-320a(miR-320a)and SRY-like high-mobility group box 4(SOX4)are closely related to tumor occurrence,progression,and prognosis.This study aims to analyze the expression of miR-320a and SOX4 in OSCC tissues and their correlation with prognosis.Methods:Tumor tissues from 62 OSCC patients(OSCC group),buccal lesion tissues from 30 patients with leukoplakia,erythroplakia,or lichen planus(precancerous lesion group),and normal oral mucosa tissues from 8 healthy volunteers(control group)were collected.The expression levels of miR-320a and SOX4 in these tissues were detected,and their correlation,as well as their relationship with clinicopathological features and overall survival(OS)of OSCC patients,were analyzed.Results:Compared to the control group,miR-320a expression levels were lower and SOX4 protien expression levels were higher in the precancerous lesion group and the OSCC group(all P<0.05).Furthermore,the OSCC group exhibited lower miR-320a levels and higher SOX4 levels compared to the precancerous lesion group(both P<0.05).MiR-320a expression levels were negatively correlated with SOX4 protein expression levels were in the OSCC patients(r=−0.282,P<0.05).Both miR-320a and SOX4 levels were associated with tumor node metastasis(TNM)staging in OSCC patients(both P<0.05).Patients with high miR-320a expression had longer OS than those with low miR-320a expression(P<0.05),while patients with high SOX4 expression had shorter OS than those with low SOX4 expression(P<0.05).High miR-320a expression was identified as a protective factor for OS in the OSCC patients(P<0.05).Conclusion:Abnormal expression of miR-320a and SOX4 may be involved in the occurrence and progression of OSCC.MiR-320a could serve as a potential prognostic biomarker for OSCC patients.
