云南嵩明大哨天然蛹虫草内生及其生境土壤可培养细菌的多样性

摘  要:了解云南嵩明大哨天然蛹虫草内生及其生境土壤中细菌物种组成、多样性等群落结构特征,丰富蛹虫草相关微生物的可利用资源多样性,增加对蛹虫草细菌区系中物种多样性的认识。采用8种分离培养基对10份来自云南嵩明大哨的蛹虫草及生境土壤样品进行细菌的分离,对部分菌株进行16S rRNA基因测序分析研究其多样性。结果表明:从蛹虫草子座、菌核及生境土壤中共分离到的373株纯培养物,对其中的217株进行测序鉴定,结果显示蛹虫草子座、菌核及生境土壤中分布有210株分属于4门21科39属87种的细菌以及7株分属于1门4科4属6种的放线菌。其中生境土壤中的细菌类群(21科27属52种)明显较子座(5科7属15种)和菌核(11科17属41种)丰富。子座和菌核中的共有优势属为芽孢杆菌属(Bacillus)和类芽孢杆菌属(Paenibacillus),共有优势种为Bacillus cereus、Bacillus mycoide和Paenibacillus terrae;生境土壤中的优势菌属为伴根瘤菌属(Pararhizobium)、贪噬菌属(Variovorax)和寡养单胞菌属(Stenotrophomonas),优势种为Pararhizobium giardinii、Variovorax sp.和Stenotrophomonas indicatrix。云南嵩明天然蛹虫草内生及其生境土壤中分布有丰富的细菌类群,这为研究蛹虫草内生及其生境土壤中细菌的生物活性积累了菌种资源。The diversity and community structure of endophytic and its habitat soil bacteria of natural Cordyceps militaris in Dashao village,Songming County,Yunnan Province were explored,which could provide basis for utilization of microbial resources and enrich our understanding of bacterial diversity in C.militaris.Ten C.militaris and its habitat soil samples were collected and isolated using 8 isolation media;16S rRNA gene sequencing was used to identify and analyze the diversity of isolated strains.A total of 373 strains were isolated,and 217 typical strains were sequenced and identified.The results showed that 210 strains belonged to 87 species,39 genera,21 families,and 4 phyla,and 7 actinobacteria belonged to 6 species,4 genera,4 families,and 1 phyla inhabited in C.militaris.The habitat soil bacterial communities(52 species,27 genera,and 21 families)were more abundant than the stroma(15 species,7 genera,and 5 families)and sclerotia(41 species,17 genera,and 11 families)in C.militaris.The dominant genera in stroma and sclerotia were Bacillus and Paenibacillus,the dominant species were Bacillus cereus,Bacillus mycoides,and Paenibacillus terrae.While the dominant genera in the habitat soil were Pararhizobium,Variovorax,and Stenotrophomonas,the dominant species were Pararhizobium giardinii,Variovorax sp.,and Stenotrophomonas indicatrix.There were abundant bacteria in the endophytic and habitat soil of natural C.militaris in Dashao village,Songming County,Yunnan Province,and the strains could be used to study the biological activities and other functions further.
