摘 要:目的:探究透明质酸(hyaluronic acid,HA)通过下调piezo蛋白缓解颞下颌关节骨关节炎(temporomandibular joint osteoarthritis,TMJOA)的潜在机制。方法:构建大鼠TMJOA模型,建模2周后予以HA治疗;HE染色、甲苯胺蓝染色、Masson染色观察TMJOA发生和HA治疗后髁突病理改变并行改良Mankin OA评分,免疫组织化学染色检测髁突软骨piezo1和piezo2的表达变化,免疫组织荧光染色检测髁突软骨GPX4的表达变化。结果:碘乙酸钠成功诱导大鼠髁突产生TMJOA样病理变化,HA治疗2周后髁突软骨损伤得到修复,改良Mankin OA评分降低;HA抑制大鼠TMJOA髁突软骨中piezo1和piezo2的高表达,同时逆转GPX4的低表达。结论:HA可能通过下调piezo1和piezo2的表达和抑制髁突软骨细胞铁死亡,进而有效缓解大鼠TMJOA的进展。Objective:To explore the potential mechanism of hyaluronic acid(HA)in alleviating temporomandibular joint osteoarthritis(TMJOA)by downregulating piezo proteins.Methods:A rat model of TMJOA was established,and HA treatment was administered two weeks after modeling.HE staining,toluidine blue staining,and Masson staining were used to observe the pathological changes of the condyle in TMJOA and after HA treatment and analyzed with a modified Mankin OA score.Immunohistochemical staining was performed to detect the expression changes of piezo1 and piezo2 in the condylar cartilage,and immunofluorescence staining was used to detect the expression changes of GPX4 in the condylar cartilage.Results:Monosodium iodacetate successfully induced TMJOA-like pathological changes in rats,and condylar cartilage damage was repaired after two weeks of HA treatment,with a decrease in the modified Mankin OA score.HA inhibited the high expression of piezo1 and piezo2 in the condylar cartilage of rats with TMJOA and simultaneously reversed the low expression of GPX4.Conclusion:HA may effectively alleviate the progression of TMJOA in rats by downregulating the expression of piezo1 and piezo2 and inhibiting ferroptosis in condylar cartilage cells.
