摘  要:基于非靶向代谢组学技术,研究马六甲肉食螨在粮仓应用后对小麦代谢物组成的差异和特点,利用四级杆-静电场轨道阱高分辨质谱联用技术分析小麦加螨和不加螨的之间的差异代谢物。结合在线和本地数据库(Thermo mzcloud)进行化学成分鉴定,采用主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)的方法进行数据模型的识别,筛选标准差异代谢物,通过MetaboAnalyst 6.0数据库分析差异代谢物代谢通路。结果表明小麦加螨和不加螨二者之间差异代谢物种类没有差异,但是物质的含量有所差异。其中鉴定到的化合物中,储藏开始时筛选得到48种差异代谢物,2个月后58种,4个月后71种,6个月后58种。加螨和不加螨经通路分析后发现主要体现在4条最主要的代谢通路上,储藏开始时富集程度最高的代谢通路为视黄醇代谢,2个月后为类固醇激素生物合成,4个月后为鞘脂代谢生物合成,均富集到1个差异代谢物,分别为视黄酸、2-氧代己二酸、生殖激素和鞘氨醇,6个月后为酪氨酸代谢,富集到2个差异代谢物,分别为假定蛋白和多球壳菌素。研究结果表明,小麦中投放马六甲肉食螨后,代谢物质种类未发生变化,物质含量产生了差异,主要是酸类和醇类,分属不同的代谢途径。本研究为应用天敌防治储粮害虫的安全性评价提供参考方法,推动天敌马六甲肉食螨防控技术的应用。Based on the non-targeted metabonomics technology,the differences and characteristics of the metabolites composition of Cheyletusmalaccensis in wheat after their application in granaries were studied,and the differential metabolites between wheat with and without mites were analyzed by quadrupole-electrostatic field orbitrap high-resolution mass spectrometry.Chemical constituents were identified by online and local databases(Thermo mzcloud).Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to identify the data model,screen the standard differential metabolites,and analyze the metabolic pathways of differential metabolites through MetaboAnalyst 6.0 database.The differential metabolites exhibited similar types but distinct content.Identified compounds yielded 48,58,71 and 58 unique metabolites after initial storage,two months,four months,and six months respectively.Pathway analysis revealed that the addition of mites was mainly reflected in the four most important metabolic pathways.The metabolic pathway with the highest degree of enrichment at the beginning of storage was retinol metabolism.After two months,it was steroid hormone biosynthesis.After four months,it was sphingolipid metabolic biosynthesis.All the above were enriched into one differential metabolite,namely retinoic acid,2-oxohexanedioic acid,testosterone,and sphingosine.After six months,tyrosine metabolism was enriched into two differential metabolites,namely putative protein and polyhedrin.The results indicated that metabolite composition remains consistent,but the substance levels exhibited changes,mainly acids and alcohols were different metabolic pathways.This study provided a reference method for the safety evaluation of using natural enemies to control stored grain pests,and promoted the application of the control technology of Cheyletusmalaccensis oudemans.
