摘 要:目的 本研究旨在识别膀胱癌(bladder cancer,BLCA)中差异表达的失巢凋亡相关基因(ARGs),构建预后模型并开发免疫评分系统,以评估患者预后及治疗效果,为膀胱癌精准治疗提供理论依据。方法 获取TCGA中膀胱癌患者的数据后识别ARGs,构建预后模型,基于预后基因开发膀胱癌免疫评分系统,分析不同组情况及探索对高风险组患者有增强治疗效果的药物。结果 根据机器学习算法,用4个关键基因(CALR、CAV1、CSPG4和NTRK2)构建了膀胱癌预后模型,构建的风险模型可将BLCA分为高风险组和低风险组,低风险组生存率更高,且该模型可作为独立预后因子,还识别出几种对高风险组患者有增强治疗效果的药物。结论 本研究提供了新的系统分析方法,用于评估膀胱癌患者的预后、临床特征、免疫细胞浸润、化疗反应和免疫治疗效果。Objective This study aims to identify anoikis-related genes(ARGs)differentially expressed in bladder cancer(BLCA),construct a prognostic model,and develop an immune scoring system to evaluate patient prognosis and treatment efficacy,providing a theoretical basis for precision therapy in BLCA.Methods After obtaining the data of bladder cancer patients in TCGA and identifying ARGs,a prognosis model was built.An immune scoring system for bladder cancer was developed based on prognostic genes to analyze the situation of different groups and identify drugs that can enhance the therapeutic effect for high-risk patients.Results A prognostic model for bladder cancer was constructed with four key genes(CALR,CAV1,CSPG4 and NTRK2)based on a machine learning algorithm.The constructed risk model divided BLCA into high-risk and low-risk groups,with a higher survival rate in the low-risk group and the model was used as an independent prognostic factor,and several drugs with an enhanced therapeutic effect on patients in the high-risk group were also identified.Conclusion This study provides new systematic analysis methods for assessing the prognosis,clinical characteristics,immune cell infiltration,chemotherapy response and immunotherapy effect in bladder cancer patients.
