摘 要:Sirtuins 1-7(SIRT1-SIRT7)是烟酰胺腺嘌呤二核苷酸依赖性去乙酰化酶家族,具有促进长寿和延缓年龄相关疾病的作用。Sirtuins通过对骨细胞的直接作用在正常的骨骼发育和体内平衡中发挥关键作用。SIRT1可以调节破骨细胞活性、促进间充质干细胞的成骨分化和骨矿化,并可以通过调节线粒体稳态从而调控骨形成和骨吸收中的骨稳态。SIRT3可以通过调节线粒体功能来调控成骨细胞分化和间充质干细胞衰老;SIRT6则可以诱导破骨细胞凋亡及促进成骨细胞活性,并且通过维持线粒体稳态来改善间充质干细胞衰老和成骨分化;而SIRT7在调控间充质干细胞成骨分化也发挥重要作用。因此Sirtuins可能是调节骨重塑和治疗骨质疏松症的潜在靶标。Sirtuins 1-7(SIRT1-SIRT7)are a family of nicotinamide adenine dinucleotide-dependent deacetylases that have the ability to promote longevity and fight age-related diseases.Sirtuins play a key role in normal bone development and homeostasis through their direct action on bone cells,and their disorder may lead to an imbalance in bone homeostasis leading to osteoporosis.SIRT1 regulates osteoclast activity,osteogenic differentiation of mesenchymal stem cells,and bone mineralization,and maintains bone homeostasis in bone formation and bone resorption by regulating mitochondrial homeostasis.SIRT3 can regulate mitochondrial function to regulate osteoblast differentiation and mesenchymal stem cell aging.SIRT6 can induce osteoclast apoptosis,promote osteoblast activity and maintain mitochondrial homeostasis to improve mesenchymal stem cell aging and osteogenic differentiation.SIRT7 also plays an important role in regulating mesenchymal stem cell osteogenic differentiation.Therefore,Sirtuins may be potential targets for regulating bone remodeling and treating osteoporosis.
