摘 要:目的通过建立抑郁症肾阳虚证模式大鼠,探究刺五加苷B对阳虚致郁大鼠的肾温煦功能的影响。方法将30只实验大鼠进行随机分组,正常组、模式组、低刺五加苷B组、中刺五加苷B组、高刺五加苷B组,每组各6只。模式组、低刺五加苷B组、中刺五加苷B组和高刺五加苷B组采用氢化可的松激素注射(腹腔注射,25 mg/kg),结合持续的不可预见的温和刺激建立肾阳虚证抑郁症大鼠模式。随后,低刺五加苷B组、中刺五加苷B组、高刺五加苷B组分别给予150 mg/kg、300 mg/kg、600 mg/kg灌胃治疗30 d。采用水迷宫、强迫游泳实验、旷场实验观察大鼠在造模及干预前后行为学的变化。使用免疫印迹法检测大鼠脑组织海马区相关基因RAP1、cAMP和RAF1的蛋白表达水平。结果干预后,与正常组比较,模式组、低刺五加苷B组、中刺五加苷B组、高刺五加苷B组大鼠的强迫游泳不动时间有所延长(P<0.01);与模式组比较,三组刺五加苷B用药组强迫游泳不动时间均缩短(P<0.01)。干预后,模式组、低刺五加苷B组、中刺五加苷B组比正常组的平台区域穿梭次数、平台潜伏期、平台象限时间占比均降低(P<0.01);与模式组比较,三组刺五加苷B用药组平台区域穿梭次数、平台潜伏期、平台象限时间占比均升高(P<0.01)。与正常组比较,模式组、低刺五加苷B组、中刺五加苷B组、高刺五加苷B组的cAMP、RAP1、RAF1表达水平均下调(P<0.01);与模式组比较,三组刺五加苷B用药组的cAMP、RAP1、RAF1蛋白表达均上调(P<0.01)。结论刺五加苷B可能通过调控cAMP-RAF1这一信号通路,来改善大鼠海马区的功能,进而改善大鼠的抑郁样行为。Objective To explore the effect of eleutheroside B on renal warming function of depression rats caused by Yang deficiency by establishing depression model rats with kidney Yang deficiency.Methods The experimental rats were randomly divided into normal group,model group,low eleutheroside B group,middle eleutheroside B group and high eleutheroside B group with 6 rats in each group.The model group,low eleutheroside B group,middle eleutheroside B group and high eleutheroside B group were injected with hydrocortisone(intraperitoneal injection,25 mg/kg)combined with continuous unpredictable mild stress to establish the depression model of kidney Yang deficiency syndrome.Then,low eleutheroside B group,middle eleutheroside B group and high eleutheroside B group were given 150 mg/kg and 300 mg/kg and 600 mg/kg gastric lavage treatment for 30 days.Water maze,forced swimming test,open field test were used to observe the behavioral changes of rats before and after modeling and intervention.Western blot method was used to detect protein expression levels of RAP1,cAMP and RAF1.Results After the intervention,compared with normal group,the forced swimming immobility time of model group,low eleutheroside B group,middle eleutheroside B group and high eleutheroside B group all prolonged(P<0.01);and compared with model group,the forced swimming immobility time of low eleutheroside B group,middle eleutheroside B group and high eleutheroside B group all shortened(P<0.01).After the intervention,compared with normal group,model group,low eleutheroside B group,middle eleutheroside B group all showed reduced shuttle times of platform area,platform incubation periods,platform quadrant time proportions(P<0.01);and compared with model group,low eleutheroside B group,middle eleutheroside B group and high eleutheroside B group all showed increased shuttle times of platform area,platform incubation periods,platform quadrant time proportions(P<0.01).Compared with normal group,the expression levels of RAP1,cAMP and RAF1 in hippocampus of model gr
