摘  要:目的探讨急性髓系白血病(AML)中miR-483-3p的表达及意义,研究其对AML细胞增殖和凋亡的影响。方法采用实时荧光定量PCR分析miR-483-3p在30例AML患者外周血单个核细胞中的表达水平,分析其与临床特征的关系。分别转染mimic和anti-miR质粒过表达及敲低AML细胞系Kasumi-1中miR-483-3p的表达,应用MTS法检测Kasumi-1细胞增殖能力变化,流式细胞术和Western blot联合分析Kasumi-1细胞凋亡水平变化。结果与正常组相比,miR-483-3p在初诊AML患者外周血单个核细胞中表达明显下调(P<0.05),miR-483-3p表达水平的高低与AML患者的年龄、性别、外周血WBC、HGB、PLT数量以及骨髓原始细胞比例、FAB分型等临床特征均无关系。过表达miR-483-3p可显著抑制Kasumi-1细胞增殖,并促进其凋亡;而敲减miR-483-3p则可显著促进其增殖,并抑制其凋亡。结论miR-483-3p在AML中发挥抑癌基因作用,并参与AML发生、发展,可作为AML潜在的标志物及新靶点。Objective To investigate the expression and significance of miR-483-3p in acute myeloid leukemia(AML)and its effect on the proliferation and apoptosis of AML cells.Methods Real-time fluorescence quantitative PCR was used to analyze the expression level of miR-483-3p in peripheral blood mononuclear cells of 30 patients with AML,and the relationship between miR-483-3p and clinical characteristics was analyzed.Separately applying transfecting mimic and anti-miR plasmid to overexpress and knock down the expression of miR-483-3p in AML cell line Kasumi-1,the proliferation ability of Kasumi-1 cells was detected by MTS method;The apoptosis level of Kasumi-1 cells was analyzed by flow cytometry and Western blot.Results Compared with the normal group,the expression of miR-483-3p in peripheral blood mononuclear cells of newly diagnosed AML patients was significantly down-regulated(P<0.05),the expression level of miR-483-3p was not correlated with the age,sex,WBC,HGB,PLT number in peripheral blood,the proportion of bone marrow original cells,FAB typing and other clinical characteristics of AML patients.Overexpression of miR-483-3p in Kasumi-1 cells significantly inhibited its proliferation and promoted its apoptosis.While knockdown of miR-483-3p significantly promoted its proliferation and inhibited its apoptosis.Conclusion miR-483-3p plays a role as a tumor suppressor gene in the occurrence and development of AML,and can be used as a potential marker and a new target for AML.
