摘  要:目的探讨STARD7在非小细胞肺癌中的表达及其对癌细胞转移活性的影响。方法使用蛋白免疫印迹实验检测BEAS-2B细胞、H358细胞、A549和H1299细胞中STARD7的表达水平。将非小细胞肺癌A549细胞分为4组:si NC组、si STARD7组、LY294002组、740Y-P组,通过MTT实验检测A549细胞的增殖活性;通过Transwell实验与划痕实验检测A549细胞的侵袭与迁移能力;通过流式细胞仪检测A549细胞的凋亡率;通过蛋白免疫印迹实验检测PI3K/AKT信号通路蛋白的表达水平。结果与BEAS-2B细胞相比,H358细胞、A549和H1299细胞的STARD7表达增加。与si NC组比较,si STARD7组与LY294002组的A549细胞的增殖活性降低,740Y-P组的A549细胞增殖活性增加(P<0.05);与si NC组比较,si STARD7组与LY294002组的A549细胞侵袭与迁移数量减少,740Y-P组的A549细胞侵袭与迁移数量增加(P<0.05);与si NC组比较,si STARD7组与LY294002组的A549细胞凋亡率增加,740Y-P组的A549细胞凋亡率降低(P<0.05);与si NC组比较,si STARD7组与LY294002组的A549细胞PI3K、AKT蛋白表达水平降低,740Y-P组的A549细胞PI3K、AKT蛋白表达水平增加(P<0.05)。结论STARD7在非小细胞肺癌中高表达。下调STARD7表达后能够抑制非小细胞肺癌A549细胞的增殖和转移,并促进A549细胞凋亡,并且STARD7的这一作用与调控PI3K/AKT通路密切相关。Objective To investigate the expression of STARD7 in non-small cell lung cancer(NSCLC)and its effect on the metastatic activity of cancer cells.Methods The expression levels of STARD7 in BEAS-2B,H358,A549 and H1299 cells were detected by Western blot.Non-small cell lung cancer A549 cells were divided into 4 groups:si NC group,si STARD7 group,LY294002 group and 740Y-P group.MTT assay was used to detect the proliferation activity of A549 cells.Transwell assay and scratch assay were used to detect the invasion and migration ability of A549 cells.The apoptosis rate of A549 cells was detected by flow cytometry.The expression levels of PI3K/AKT signaling pathway proteins were detected by Western blot.Results Compared with BEAS-2B cells,the expression of STARD7 was increased in H358,A549 and H1299 cells.Compared with the si NC group,the proliferation activity of A549 cells in the si STARD7 group and LY294002 group was significantly decreased,and the proliferation activity of A549 cells in the 740Y-P group was significantly increased(P<0.05).The number of invasion and migration of A549 cells in the si STARD7 and LY294002 groups was significantly decreased,while the number of invasion and migration of A549 cells in the 740Y-P group was significantly increased(P<0.05).The apoptosis rate of A549 cells was increased in the si STARD7 group and LY294002 group,and decreased in the 740Y-P group(P<0.05).The protein expression levels of PI3K and AKT in A549 cells were decreased in the si STARD7 group and LY294002 group,and increased in the 740Y-P group(P<0.05).Conclusion STARD7 is highly expressed in non-small cell lung cancer.Down-regulation of STARD7 expression can inhibit the proliferation and metastasis of A549 cells,and promote the apoptosis of A549 cells.The effect of STARD7 is closely related to the regulation of PI3K/AKT pathway.
