摘  要:为探索羧化壳聚糖(carboxymethyl chitin, CMCS)在猪精液常温保存过程中的抗氧化及抗菌作用,采集某猪场杜洛克品系公猪不同批次精液,并在常温条件下保存,第0天和第3天添加3种浓度CMCS(0.05 mg·mL^(-1)、0.10 mg·mL^(-1)、0.15 mg·mL^(-1)),分别测定其对精子活力、总抗氧化能力(T-AOC)、活性氧水平(ROS)、丙二醛浓度(MDA)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)活性影响,筛选确定CMCS的最佳添加浓度;同时对所有批次精液进行细菌分离鉴定并测定CMCS抗菌效果。结果表明,与空白对照相比,在常温保存过程中,添加0.05 mg·mL^(-1)CMCS可显著提高精子活力及TAOC、CAT、SOD、GSH-PX抗氧化活性(P<0.05),降低ROS和MDA氧化指标(P<0.05)。另外,从采集的精液中分离出3株革兰氏阴性细菌,16SrDNA测序分析结果显示,3株细菌与变形杆菌参考菌株相似性最高,分别为99.93%、100%、99.79%,并且0.05mg·mL^(-1)壳聚糖添加组可显著降低猪精液常温保存中的细菌含量(P<0.05)。综上,CMCS在猪精液常温保存过程中的适宜添加浓度为0.05 mg·mL^(-1),该浓度CMCS可有效提高猪精液常温保存过程中的抗氧化性及抗菌活性。研究结果可为CMCS在猪精液常温保存过程中的应用提供理论依据。To explore the antioxidant and antibacterial effects of carboxymethyl chitosan(CMCS)in boar semen during normal temperature,different batches of Duroc male boar semen were collected from pig farms.The total antioxidant capacity(T-AOC),reactive oxygen species(ROS),malondialdehyde(MDA)content,catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GSH-PX)activity were determined on day 0 and day 3 under room temperature storage conditions in three CMCS concentrations(0.05 mg·mL^(-1),0.10 mg·mL^(-1),0.15 mg·mL^(-1))groups for determination the optimal concentration.Simultaneously,bacterial isolation and identification were performed on all batches of semen,and the antibacterial effect of CMCS was further analyzed.The results showed that compared with the blank control,adding 0.05 mg·mL^(-1) CMCS could significantly increase sperm motility and antioxidant activities such as T-AOC,CAT,SOD,GSH-PX(P<0.05),while it could reduce oxidative indicators such as ROS and MDA(P<0.05)during room temperature storage,suggestting that 0.05 mg·mL^(-1)CMCS was the appropriate concentration inboar semen.Three strains of Gram negative bacteria were isolated from some batches of semen,and the 16S rDNA sequencing analysis showed that the bacteria were highest similarity with the reference strain of Proteus,with 99.93%,100%,and 99.79%,respectively.Moreover,0.05 mg·mL^(-1) CMCS could significantly reduce the bacterial content during room temperature storage(P<0.05).In conclusion,the appropriate concentration of CMCS in normal temperature storage of pigs is 0.05 mg·mL^(-1),which can effectively improve the antioxidant and antibacterial activities of pigs in normal temperature storage.The results provide a theoretical basis for the application of CMCS in normal temperature storage of pigs.
