微生物发酵制备红景天香原料及其特征成分分析

摘　　要：[目的]利用酿酒酵母发酵制备红景天香原料,并分析其特征成分。[方法]红景天粉末经超声提取、灭活、接种酿酒酵母种子液、发酵、一次醇提和过滤、二次醇提和过滤、浓缩等步骤得到天然香原料。建立UPLC-MS/MS方法对该香原料中红景天苷、酪醇、络塞维、红景天素、槲皮素5种成分进行定量分析,探讨酿酒酵母糖基转移酶(UGT)在红景天苷合成中的作用。[结果]制备所得红景天香原料具有浓郁芳香气味,与对照样品相比,红景天苷含量提高了14.8%,酪醇含量提高了11.7%,络塞维、红景天素、槲皮素含量增加不明显。[结论]该研究利用微生物发酵技术结合化学成分检测和qPCR技术,制备了安全稳定、活性成分含量较高的天然香原料,为其在食品添加剂、烟草、日化等领域的应用提供理论依据。[Objective]To prepare Rhodiola perfume by fermentation with Saccharomyces cerevisiae and analyze its characteristic components.[Method]Rhodiola powder underwent ultrasonic extraction,inactivation treatment,inoculation of Saccharomyces cerevisiae seed solution,fermentation,primary alcohol extraction and filtration,secondary alcohol extraction and filtration,concentration,and other steps to obtain natural aroma raw materials.UPLC-MS/MS method was established to quantitatively analyze the five components of salidroside,tyrosol,rosavin,rhodiosin and quercetin in the raw materials.The role of Saccharomyces cerevisiae glycosyltransferase(UGT)in salidroside synthesis was investigated.[Result]The prepared Rhodiola rosea perfume had a strong aromatic aroma.Compared with the control sample,the content of salidroside increased by 14.8%,the tyrosol content increased by 11.7%,and the content of rosavin,rhodiosin and quercetin did not increase significantly.[Conclusion]The microbial fermentation technology combined with chemical composition detection and qPCR technology were used to prepare safe and stable natural aroma raw materials with high content of active ingredients,which provided theoretical basis for their application in food additives,tobacco,daily chemical and other fields.
