摘  要:目的基于蛋白酪氨酸激酶2/信号转导与激活因子3/环氧合酶-2(JAK2/STAT3/COX-2)信号通路探讨偏瘫口服液对缺血性脑卒中大鼠脑损伤的保护作用。方法取SPF级8周龄SD大鼠40只,随机选择6只作为正常组,其余34只大鼠利用改良Longa线栓法构建缺血性脑卒中模型。将建模成功的大鼠随机分为模型组、偏瘫口服液组、白细胞介素-6(IL-6)组、偏瘫口服液+IL-6组、丁苯酞组,每组6只。偏瘫口服液组给予偏瘫口服液6.0 g/kg灌胃,IL-6组给予IL-6抗体0.5 mg/kg尾静脉注射,偏瘫口服液+IL-6组给予偏瘫口服液6.0 g/kg灌胃和IL-6抗体0.5 mg/kg尾静脉注射,丁苯酞组给予丁苯酞软胶囊70 mg/kg灌胃,正常组、模型组给予等体积生理盐水灌胃,均1次/d。连续干预7 d后,记录各组大鼠神经功能评分,计算脑梗死体积比,HE染色观察脑组织病理形态,比色法检测血清氧化应激指标超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、丙二醛(MDA)水平,酶联免疫吸附法检测血清炎症因子白细胞介素-1β(IL-1β)、IL-6、肿瘤坏死因子-α(TNF-α)水平,RT-qPCR法和Western blot法分别检测脑组织中JAK2、STAT3、COX-2、血管内皮生长因子(VEGF)mRNA和蛋白表达情况。结果与正常组比较,模型组和IL-6组大鼠脑组织出现典型的缺血性损伤,神经功能评分、脑梗死体积比和血清MDA、IL-1β、IL-6、TNF-α水平及脑组织中JAK2、STAT3、COX-2 mRNA和蛋白相对表达量均明显升高(P均<0.05),血清SOD、CAT水平及脑组织中VEGF mRNA和蛋白相对表达量均明显降低(P均<0.05),且IL-6组上述指标变化较模型组更明显(P均<0.05)。与模型组比较,偏瘫口服液组、丁苯酞组、偏瘫口服液+IL-6组大鼠脑组织损伤明显减轻,神经功能评分、脑梗死体积比和血清MDA、IL-1β、IL-6、TNF-α水平及脑组织中JAK2、STAT3、COX-2 mRNA和蛋白相对表达量均明显降低(P均<0.05),血清SOD、CAT水平及脑组织中VEGF mRNA和蛋白相对表达�Objective It is to investigate the protection effect of Hemiplegic oral solution(HOS)in rats with ischemic stroke based on Janus tyrosine kinase 2(JAK2)/signal transducer and activator of tranion 3(STAT3)/COX-2.Methods Totally 408-week-old SD rats of SPF grade were selected,6 of them were taken as normal group,and the other 34 rats were used to establish models of ischemic stroke with modified Longa suture method.The successfully modeled rats were randomly divided into model group,HOS group,interleukin-6(IL-6)group,HOS+IL-6 group and butylphthalide group,with 6 rats in each group.The HOS group was given HOS 6.0 g/kg by gavage,the IL-6 group was given IL-6 antibody 0.5 mg/kg by caudal vein injection,the HOS+IL-6 group was given HOS 6.0 g/kg by gavage and IL-6 antibody 0.5 mg/kg by caudal vein injection,the butylphthalide group was given butylphthalide soft gelatin capsule 70 mg/kg by gavage,and the normal group and model group were given equal volume of normal saline by gavage,all once daily.After 7 days of continuous intervention,the neurological function scores of rats in each group were recorded,and the volume ratio of cerebral infarction was calculated,the pathological morphology of brain tissues was observed by HE staining,the levels of serum oxidative stress indexes such as superoxide dismutase(SOD),catalase(CAT)and malondialdehyde(MDA)were detected by colorimetric assay,the serum levels of inflammatory factors such as interleukin-1β(IL-1β),IL-6 and tumor necrosis factor-α(TNF-α)were detected by ELISA,the mRNA and protein expressions of JAK2,STAT3,COX-2 and vascular endothelial growth factor(VEGF)in brain tissues were detected by RT-qPCR and Western blot,respectively.Results Compared with the normal group,the brain tissues of rats in the model group and IL-6 group showed typical ischemic injury,and the neurological function scores,cerebral infarction volume ratio,and serum levels of MDA,IL-1β,IL-6 and TNF-α,as well as the relative expressions of JAK2,STAT3 and COX-2 mRNA and proteins in brain tissues w
