摘  要:建立测定凤丹皮滴丸中芍药苷的HPLC方法,并对其体外抗氧化性进行初步研究。采用HPLC法测定,色谱条件选用依利特C 18柱(4.6 mm×250 mm,5μm),流动相为乙腈∶0.1%磷酸(14∶86),检测波长为230 nm,进样体积为10μL,流速为1.0 mL·min^(-1),柱温为30℃。同时采用UV-vis法测定,取芍药苷、Vc溶液分别与1,1-二苯基-2-三硝基苯肼溶液(DPPH)等体积混合,用50%乙醇为空白,Vc为对照,暗处放置,于517 nm波长处测定吸光度。在1.786~17.86μg·mL^(-1)浓度区间范围内,芍药苷表现出良好的线性关系,r=0.9998,线形回归方程为y=10.338 x-2.0567,RSD=1.5%,平均回收率为95.4%(n=6)。凤丹皮滴丸中芍药苷的使DPPH自由基的吸收率达到50%所需的浓度为31.28μg·mL^(-1),表明芍药苷对DPPH自由基的清除作用较好。本试验方法准确可靠,灵敏度高、操作简单、重现性好,可用于芍药苷的定量分析,同时在体外抗氧化性方面,凤丹皮滴丸中芍药苷表现出良好的特性。The method of HPLC was established for the determination of paeoniflorin in the dripping pills of Cortex Moutan and to conduct a preliminary study on its in vitro antioxidant activity.The method of HPLC was used for determination,and the chromatographic conditions were selected using YiliTe C 18 column(4.6 mm×250 mm,5μm).The mobile phase was acetonitrile∶0.1%phosphoric acid(14∶86),with the detection wavelength of 230 nm,the injection volume of 10μL,the flow rate of 1.0 mL·min^(-1),and the column temperature of 30℃.Simultaneously,the method of UV vis was used for determination.Paeoniflorin and Vc solutions were mixed with equal volumes of 1,1-diphenyl-2-trinitrophenylhydrazine solution(DPPH),and 50%ethanol was used as the blank and the control of Vc.The absorbance was measured at the wavelength of 517 nm in the dark.In the concentration range of 1.786 to 17.86μg·mL^(-1),paeoniflorin showed good linear relationship,with r=0.9998.The linear regression equation was y=10.338 x-2.0567,RSD=1.5%,and the average recovery rate was 95.4%(n=6).The concentration required for the absorption rate of DPPH free radicals by paeoniflorin in the dripping pills of Cortex Moutan to reach 50%was 31.28μg·mL^(-1),indicating that paeoniflorin had the good scavenging effect on DPPH free radicals.The experimental method is accurate,reliable,easy to operate,highly sensitive,and has good reproducibility.It can be used for quantitative analysis of paeoniflorin.At the same time,in terms of in vitro antioxidant activity,paeoniflorin in the dripping pills of Cortex Moutan exhibits good characteristics.
