摘 要:以琥珀蚕(Antheraea assamensis)茧为原料,采用碱溶酸沉法提纯制备琥珀蚕茧黑色素。对分离得到的黑色素样品进行溶解性试验和稳定性试验,采用紫外可见光谱和傅里叶红外光谱法分析琥珀蚕茧黑色素的结构特性,检测样品的总抗氧化能力及ABTS、DPPH、PTIO自由基清除能力,以评价其体外抗氧化能力。结果表明,琥珀蚕茧黑色素呈黑棕色粉末,得率约为0.03%,色价为428.59,溶于碱性溶液,不溶于水及其他常见有机溶剂;其紫外吸收光谱与红外光谱具有吲哚型黑色素典型光谱特征;琥珀蚕茧黑色素具有良好的光热稳定性,对氧化剂和还原剂耐受性不强;体外抗氧化实验表明琥珀蚕茧黑色素具有一定的抗氧化能力,并且在试验范围内低于黑色素标准品。这为研究黑色素在琥珀蚕茧丝中的生理功能奠定了基础。The melanin derived from the cocoon of Antheraea assamensis was isolated and purified by the method of alkali-dissolution and acid-isolation.The isolated melanin was evaluated by solubility test and stability test,and then the structural characteristics was studied by ultraviolet-visible spectroscopy(UV-Vis)and Fourier transform infrared spectroscopy(FTIR).The total antioxidant capacity and ABTS,DPPH and PTIO radical scavenging activities were tested to evaluate the in vitro antioxidant activity of the melanin.The result showed that the melanin powder was black brown.The yield was about 0.03%,and the color valence was 428.59.The solubility showed that the melanin was soluble in alkaline solution,but insoluble in water and other common organic solvent.It may be a type of indoles melanin from the UV-visible and infrared spectrogram.The melanin has excellent photothermal stability but poor stability to oxidant and reducing agent.The results of in vitro antioxidant testing showed that the melanin had certain antioxidant activity,but lower than that of the melanin standard.This study lay foundations for further research on physiological functions of melanin in silk of Antheraea assamensis.
