摘  要:目的拟采用复合高脂蛋氨酸-胆碱缺乏(methionine-choline-deficient,MCD)饮食诱导小鼠建立高质量的非酒精性脂肪性肝炎(nonalcoholic steatohepatitis,NASH)模型。方法取健康雄性小鼠48只,按其饲养周期不同随机分为4、6、8周正常对照和模型6组:分别采用高脂蛋氨酸-胆碱缺乏饮食(high-fatty methionine-choline-deficient diet,HFMRCD)和正常饲料喂养后,称重记录处死取血,动态观察其饲养期末体质量及肝湿重变化、肝脏形态、病理变化及炎症因子水平。结果模型组小鼠体质量低于正常组(P<0.05);第4周模型组肝脏出现色泽变淡、质脆、触之有油腻感、体积变大呈脂肪肝状;镜下观察模型组小鼠肝脏脂变程度由第4周的70%进展至第8周的90%,其中肝小叶可见大量炎细胞呈灶状聚集,且坏死灶明显超过5个(P<0.05);模型组小鼠肝组织IL-6、TNF-α、TGF-β1含量均明显高于正常组(P<0.05)。结论复合高脂MCD饲料饮食诱导下成功建立C57BL6小鼠NASH模型。Objective To establish a high-quality model of nonalcoholic steatohepatitis(NASH)using compound high-fat methionine-choline-deficient(MCD)diet induced mice.Methods Forty-eight healthy male mice were randomly divided into 6 groups of normal control and model at 4,6 and 8 weeks according to their feeding cycles:after being fed with high-fatty methionine-choline-deficient diet(HFMRCD)and normal diet,respectively.Blood was extracted by weighing and recording execution,and the changes in body weight and liver wet weight,liver morphology,pathological changes and inflammatory factor levels at the end of their feeding period were dynamically observed.Results The body weight of the model group was lower than that of the normal group(P<0.05).At the 4th week,the color of the model group became lighter,larger and showed fatty liver shape.In the model group,the degree of liver lipids improved from 70%at the 4th week to 90%at the 8th week,in which a large number of inflammatory cells were concentrated in the interstitial tissue,and there were more than 5 necrotic lesions(P<0.05),which were consistent with the characteristics of NASH.The levels of IL-6,TNF-αand TGF-β1 in the liver tissue of the model group were significantly higher than those in the normal group(P<0.05).Conclusion A NASH model of C57BL/6 mice was successfully established under the induction of compound high-fat MCD diet.
