摘 要:目的研究口乳杆菌BE7N(Lactobacillus oris BE7N,L.oris BE7N)对db/db小鼠脂代谢的影响。方法将db/db小鼠随机分为空白对照组和L.oris BE7N干预组,灌胃处理4周,期间监测体质量、摄食量和肛温情况。称重肝脏和脂肪组织;HE染色观察肝脏和棕色脂肪组织形态改变;检测血清中TC、TG、LDL-C、HDL-C水平;RT-PCR检测棕色脂肪组织中PPARγ、PGC-1α、UCP1、PRDM16、ELOVL3 mRNA的表达。结果与空白对照组相比,L.oris BE7N干预可显著延缓db/db小鼠体质量增加(P<0.01),提高小鼠核心体温(P<0.05),降低肝脏和内脏脂肪组织湿重(P<0.05),降低外周血中TC和LDL-C含量(P<0.05);减轻肝脏和棕色脂肪组织的脂质沉积;L.oris BE7N干预显著提高db/db小鼠棕色脂肪组织产热相关基因PPARγ、PGC-1α、UCP1、PRDM16和ELOVL3的mRNA表达水平(P<0.05)。结论L.oris BE7N可以通过调节棕色脂肪组织PPARγ/PGC-1α/UCP1信号通路,减轻脂肪沉积,改善db/db小鼠脂代谢异常。Objective To investigate the effects of Lactobacillus oris BE7N(L.oris BE7N)on lipid metabolism in db/db mice.Methods Db/db mice were randomly divided into control group and L.oris BE7N intervention group.Both groups received oral gavage for 4 weeks.Body weight,food intake,and rectal temperature were monitored during the study.Liverand adipose tissues were weighed.Morphology changes in the liver and brown adipose tissue were assessed using HE staining.Serum levels of TC,TG,LDL-C,and HDL-C were measured and the levels of PPARγ,PGC-1α,UCP1,PRDM16 and ELOVL3 mRNA in brown adipose tissue were detected by RT-PCR.Results Compared with the control group,L.oris BE7N intervention significantly slowed body weight gain in db/db mice(P<0.01),increased core body temperature(P<0.05),reduced liver and visceral fat mass(P<0.05),and decreased serum TC and LDL-C levels(P<0.05).Lipid deposition in the liver and brown adipose tissue was alleviated,and L.oris BE7N significantly increased the levels of thermogenesis-related genes,including PPARγ,PGC-1α,UCP1,PRDM16 and ELOVL3 in brown adipose tissue(P<0.05).Conclusion L.oris BE7N can improve lipid metabolism disorders and reduce fat deposition in db/db mice by regulating the PPARγ/PGC-1α/UCP1 signaling pathway in brown adipose tissue.
