褐藻素治疗肝纤维化的作用机制分析

摘  要:目的分析褐藻素(FX)治疗肝纤维化的作用机制。方法选择LX-2细胞,分为空白组(n=40)、低剂量组(n=40)、高剂量组(n=40),低剂量组、高剂量组分别给予15、30μmol/L FX处理,比较三组细胞增殖、凋亡、活化及细胞外基质(ECM)等,比较三组细胞Janus激酶/信号转导与转录激活子(JAK/STAT)通道改变。选择30只SPF级雄性Wistar大鼠,均注射四氯化碳(CCl4)建立肝纤维化大鼠模型,分为模型组、FX组,FX组给予250 mg/kg FX灌胃处理,比较两组大鼠肝组织纤维化改变及肝功能变化。结果低剂量组、高剂量组LX-2细胞增殖活力低于空白组,凋亡率高于空白组,高剂量组LX-2细胞增殖活力低于低剂量组,凋亡率高于低剂量组,差异有统计学意义(P<0.05)。低剂量组、高剂量组LX-2细胞α-平滑肌肌动蛋白(α-SMA)、间质胶原蛋白(COL)-Ⅰ水平低于空白组,高剂量组LX-2细胞α-SMA、COL-Ⅰ水平低于低剂量组,差异有统计学意义(P<0.05)。低剂量组、高剂量组LX-2细胞JAK蛋白、STAT蛋白水平低于空白组,高剂量组LX-2细胞JAK蛋白、STAT蛋白水平低于低剂量组,差异有统计学意义(P<0.05)。Masson染色结果显示,模型组大鼠肝组织细胞形态多为不规则状态,可见炎症细胞浸润,肝组织呈纤维化改变,FX组大鼠肝组织纤维增生症状改善。FX组血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)水平低于模型组,差异有统计学意义(P<0.05)。结论FX可通过介导JAK/STAT通路抑制LX-2细胞增殖及活化、促进LX-2细胞凋亡等生物学活动,可帮助延缓大鼠肝脏纤维化,改善肝功能,FX可作为治疗肝纤维化的一种新思路。Objective To analyze the fucoxanthin(FX)in treating hepatic fibrosis.Methods LX-2 cells were selected and divided into blank group(n=40),low-dose group(n=40)and high-dose group(n=40).The low-dose group and high-dose group were treated with 15 and 30μmol/L FX,respectively,and the proliferation,apoptosis,activation and extracellular matrix(ECM)of the three groups were compared.Janus kinase and signal transduction and transcription activator(JAK/STAT)channel changes were compared in three groups of cells.Thirty SPF male Wistar rats were selected and injected with carbon tetrachloride(CCl4)to establish a rate model of hepatic fibrosis.The rats were divided into model group and FX group.The FX group was given 250 mg/kg FX by gavage,and the changes of liver fibrosis and liver function were compared between the two groups.Results The proliferation activity of LX-2 cells in low-dose group and high-dose group were lower than those in blank group,and the apoptosis rates were higher than those in blank group,the proliferation activity of LX-2 cells in high-dose group was lower than that in low-dose group,and the apoptosis rate was higher than that in low-dose group,with statistical significances(P<0.05).The levels ofα-smooth muscle actin(α-SMA)and interstitial collagen(COL)-Ⅰin LX-2 cells in low-dose group and high-dose group were lower than those in blank group,and the levels ofα-SMA and COL-Ⅰin LX-2 cells in high-dose group were lower than those in low-dose group,with statistical significances(P<0.05).The JAK protein and STAT protein levels of LX-2 cells in low-dose group and high-dose group were lower than those in blank group,and the JAK protein and STAT protein levels of LX-2 cells in high-dose group were lower than those in low-dose group,the differences were statistically significant(P<0.05).Masson staining results showed that the morphology of liver tissue cells in the model group was mostly irregular,with inflammatory cell infiltration and fibrosis changes in the liver tissue.The symptoms of liver tissue fi
