电针联合薯蓣皂苷元对脑缺血再灌注损伤大鼠黑色素瘤缺乏因子2炎症小体介导的细胞焦亡的影响※

摘  要:目的观察电针联合薯蓣皂苷元对脑缺血再灌注损伤大鼠黑色素瘤缺乏因子2(AIM2)炎症小体介导的细胞焦亡的影响。方法将50只大鼠随机分为假手术组、模型组、电针组、薯蓣皂苷元组、联合组,每组10只。比较各组大鼠神经功能评分;酶联免疫吸附法检测脑组织中白细胞介素1β(IL-1β)、IL-6、肿瘤坏死因子α(TNF-α)水平;红四氮唑(TTC)染色法检测脑组织梗死面积;苏木素-伊红(HE)染色观察脑组织病理学变化;原位末端标记法(TUNEL)检测神经细胞凋亡率;蛋白免疫印迹法检测脑组织中AIM2、半胱天冬酶1(caspase-1)蛋白表达;荧光定量聚合酶链式反应(RT-PCR)检测脑组织中AIM2、caspase-1 mRNA表达。结果与假手术组比较,模型组大鼠神经功能评分、脑组织中IL-1β、IL-6、TNF-α水平、脑梗死面积、神经细胞凋亡率、脑组织中AIM2和caspase-1蛋白及mRNA表达均明显增加(P<0.05);与模型组比较,电针组、薯蓣皂苷元组、联合组大鼠神经功能评分、脑组织中IL-1β、IL-6、TNF-α水平、脑梗死面积、神经细胞凋亡率、脑组织中AIM2和caspase-1蛋白及mRNA表达均明显降低(P<0.05);且联合组大鼠神经功能评分、脑组织中IL-1β、IL-6、TNF-α水平、脑梗死面积、神经细胞凋亡率、脑组织中AIM2和caspase-1蛋白及mRNA表达明显低于电针组、薯蓣皂苷元组(P<0.05)。结论电针联合薯蓣皂苷元可抑制脑缺血再灌注损伤大鼠AIM2炎症小体介导的细胞焦亡。Objective To investigate the effect of electroacupuncture(EA)combined with diosgenin element on absent in melanoma 2(AIM2)inflammasome-mediated pyroptosis in rats with cerebral ischemia-reperfusion injury(CIRI).Methods Rats were randomly divided into sham operation group(sham group),CIRI group,EA group,diosgenin element group(DG group),and EA combined with diosgenin element group(EA+DG group),with 10 rats in each group.The neurological function scores of rats in each group were compared,and the levels of interleukin 1β(IL-1β),IL-6 and tumor necrosis factor alpha(TNF-α)in brain tissue were detected by enzyme-linked immunosorbent assay(ELISA).The infarct area in brain tissue was detected by triphenyl tetrazolium chloride(TTC)staining.The pathological changes in brain tissue were detected by hematoxylin and eosin(H&E)staining.The apoptosis rate of neuronal cells was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL).Protein and mRNA expressions of AIM2 and caspase-1 in brain tissue were detected by Western blotting and reverse transcription polymerase chain reaction(RT-PCR),respectively.Results Compared with the sham group,rats in the CIRI group had significantly higher neurological function scores,IL-1β,IL-6 and TNF-αlevels in brain tissue,brain infarct area,neuronal apoptosis rate and mRNA and protein expressions of AIM2 and caspase-1(P<0.05).Compared with the CIRI group,rats in the EA,DG and EA+DG groups had significantly lower neurological function score,IL-1β,IL-6 and TNF-αlevels in brain tissues,brain infarct area,apoptosis rate of neuronal cells and mRNA and protein expressions of AIM2 and caspase-1(P<0.05).The neurological function score,IL-1β,IL-6,TNF-αlevels in brain tissues,brain infarct area,apoptosis rate of neuronal cells and mRNA and protein expressions of AIM2 caspase-1 in cells were significantly lower in the EA+DG group than those of EA and DG groups(P<0.05).Conclusion EA combined with diosgenin element inhibits AIM2 inflammasome-mediated pyroptosis in
