摘　　要：目的 探讨微小RNA-106b-25(miR-106b-25)簇(miR-106b、miR-93和miR-25)在非小细胞肺癌(NSCLC)组织中的表达水平并分析其在NSCLC诊断和预后中的价值。方法 收集NSCLC患者的癌组织及癌旁正常组织60对,通过实时定量PCR检测上述组织中miR-106b-25簇的表达水平,并分析其表达水平与临床病理参数间的相关性及临床应用价值。结果 NSCLC患者组织中miR-106b和miR-93的表达水平高于对照组,且miR-106b-25簇表达水平与患者的肿瘤大小、分化程度、淋巴结转移、远处转移和TNM分期显著相关(P<0.05);miR-106b、miR-93和miR-25诊断NSCLC的受试者工作特征曲线下面积分别为0.759、0.786和0.746,三者联合检测的曲线下面积则为0.826,显著高于细胞角蛋白19片段的0.717。miR-106b-25簇中高表达miR-106b患者的5年生存率低于低表达患者(P=0.043)。结论 miR-106b-25簇是NSCLC的潜在诊断指标,其中miR-106b是有效的预后因子,对提高NSCLC患者预后预测水平具有重要意义。Objective To identify the expression level of microRNA-106b-25(miR-106b-25)cluster(miR-106b,miR-93 and miR-25)and further investigate its role in diagnosis and prognosis of non-small cell lung cancer(NSCLC).Methods Expression levels of miR-106b-25 were detected by qPCR in 60 primary tissue samples collected from patients with NSCLC,and the association between clinical characteristics and expression levels of miR-106b-25 cluster was also analyzed.Results Both miR-106b and miR-93 expression levels were significantly increased in primary tissues of NSCLC patients.Additionally,expression levels of miR-106b-25 cluster were positively correlated with tumor size,degree of differentiation,lymph node metastases,distant metastasis and TNM stage(P<0.05).The area under the curve of miR-106b,miR-93 and miR-25 were 0.759,0.816 and 0.746,respectively.Moreover,the area under receiver operating characteristc curve of miR-106b-25 cluster was 0.826,significant higher than 0.717 of cytokeratin 19 fragment in the diagnosis of NSCLC.Kaplan-Meier survival analysis indicated that patients with high expression of miR-106b was associated with poor 5-year overall survival rate compared with patients with low expression of miR-106b(P=0.043).Conclusion MiR-106b-25 cluster express at a relative high level and miR-106b is a prognostic indicator for NSCLC patients.Thus,we believe that the miR-106b-25 cluster can be an important indicator for diagnosis and prognosis of NSCLC patients.
