摘  要:目的 探讨乳腺癌组织中蛋白激酶N1(PKN1)和蛋白激酶N2(PKN2)的水平及临床意义。方法 采用实时定量PCR检测94对乳腺癌组织和癌旁组织中的PKN1、PKN2水平,结合临床病理特征分析两者在乳腺癌中的临床意义,借助Kaplan-Meier Plotter数据库在线分析两者对乳腺癌预后的影响。结果 乳腺癌组织的PKN1和PKN2水平分别4.804±1.750和4.376±1.611,均高于癌旁组织的1.743±0.912和1.670±0.850,差异均有统计学意义(t=15.045、14.402,P<0.001),Pearson相关分析显示乳腺癌组织的PKN1和PKN2水平呈正相关(r=0.605,P<0.001);分层分析发现组织PKN1水平与肿瘤大小(r=0.209,P=0.038)、淋巴结转移(r=0.200,P=0.048)、TNM分期(r=0.304,P=0.002)、组织学分级(r=0.313,P=0.001)和诺丁汉预后指数(r=0.297,P=0.011)呈正相关。PKN2水平与肿瘤大小(r=0.297,P=0.003)、淋巴结转移(r=0.309,P=0.002)和TNM分期(r=0.430,P<0.001)呈正相关。Kaplan-Meier Plotter在线分析显示,PKN1与乳腺癌的无复发生存期(HR=0.85,95%CI:0.77~0.94)和总生存期(HR=0.79,95%CI:0.65~0.95)有关,而PKN2仅与乳腺癌的无复发生存期(HR=0.77,95%CI:0.70~0.85)有关。结论 PKN1、PKN2在乳腺癌中高表达,参与了乳腺癌的恶性进展且均与复发有关,有望成为乳腺癌防治的潜在靶点。Objective To investigate the levels and clinical significance of protein kinase N1(PKN1)and protein kinase N2(PKN2)in breast cancer tissues.Methods Real-time quantitative PCR(qPCR)was used to detect the levels of PKN1 and PKN2 in 94 pairs of breast cancer tissues and adjacent tissues.Clinical significance of both levels in breast cancer were investigated using clinical and pathological characteristics,and the impact of both levels on the prognosis of breast cancer was analyzed online with the help of Kaplan-Meier Plotter database.Results The levels of PKN1 and PKN2 in breast cancer tissues were 4.804±1.750 and 4.376±1.611,higher than 1.743±0.912 and 1.670±0.850 in adjacent tissues,and the difference was statistically significant(t=15.045,14.402,P<0.001).Pearson correlation analysis showed that the levels of PKN1 and PKN2 in breast cancer tissues were positively correlated(r=0.605,P<0.001).Stratified analysis revealed a positive correlation between tissue PKN1 level and tumor size(r=0.209,P=0.038),lymph node metastasis(r=0.200,P=0.048),TNM staging(r=0.304,P=0.002),histological grading(r=0.313,P=0.001),and Nottingham prognostic index(r=0.297,P=0.011).The level of PKN2 was positively correlated with tumor size(r=0.297,P=0.003),lymph node metastasis(r=0.309,P=0.002),and TNM staging(r=0.430,P<0.001).Kaplan-Meier Plotter online analysis showed that PKN1 was related to the recurrence free survival(HR=0.85,95%CI:0.77-0.94)and overall survival(HR=0.79,95%CI:0.65-0.95)of breast cancer,while PKN2 was only related to the recurrence free survival(HR=0.77,95%CI:0.70-0.85)of breast cancer.Conclusion PKN1 and PKN2 are highly expressed in breast cancer,participate in the malignant progression of breast cancer and are related to recurrence.Both are expected to become potential targets for prevention and treatment of breast cancer.
