摘　　要：目的 探讨AT富交互域5A(ARID5A)在宫颈癌组织中的表达及其与预后的关系。方法 利用生物信息学和免疫组化En Vision两步法分析宫颈癌组织中ARID5A的表达,及其与临床病理特征、预后和免疫细胞浸润水平的关系;基因富集分析ARID5A可能参与的通路。结果 生物信息学分析显示,ARID5A在宫颈癌组织中的表达低于癌旁组织(P<0.001),且ARID5A低表达者的总生存期短于高表达者(P=0.030);免疫组化证实在宫颈癌组织中ARID5A蛋白低表达率为67.50%(54/80),高于癌旁组织的23.75%(19/80)(P=0.007);临床病理特征分析显示,ARID5A表达与宫颈癌患者的FIGO分期、淋巴结转移、脉管瘤栓、分化程度、Ki67和P16表达相关(P<0.05);生存分析显示,ARID5A低表达者的总生存期和无病生存期明显短于高表达者(P<0.05);Cox回归分析发现,ARID5A低表达和低分化是影响宫颈癌患者预后的独立危险因素(P<0.05);免疫细胞浸润分析显示,宫颈癌组织中ARID5A的表达与NK细胞、巨噬细胞和B细胞浸润水平呈正相关;基因富集分析显示,ARID5A可能参与体液免疫反应、细胞因子细胞因子受体相互作用、核受体和过氧化物酶体增殖物激活受体信号通路。结论 ARID5A在宫颈癌中低表达,且ARID5A低表达是影响宫颈癌患者预后的独立危险因素,有望作为宫颈癌预后评估的分子标志物。Objective To investigate the expression of AT rich interactive domain 5A(ARID5A)in cervical cancer tissues and its relationship with prognosis.Methods Bioinformatics and immunohistochemical EnVision two-step method were used to analyze the expression of ARID5A in cervical cancer tissues and its relationship with clinicopathological features,prognosis and immune cell infiltration level.Gene enrichment analysis of possible pathways involved in ARID5A was performed.Results Bioinformatics analysis showed that the ARID5A expression in cervical cancer tissues was lower than that in adjacent tissues(P<0.001),and the overall survival of patients with low ARID5A expression was shorter than that of patients with high ARID5A expression(P=0.030).Immunohistochemistry confirmed that the low expression rate of ARID5A protein in cervical cancer tissues was 67.50%(54/80),higher than 23.75%(19/80)in adjacent tissues(P=0.007).Clinicopathological analysis showed that the ARID5A expression was correlated with FIGO stage,lymph node metastasis,vascular thrombus,differentiation degree,Ki67 and P16 expression in cervical cancer patients(P<0.05).Survival analysis showed that the overall survival and disease-free survival in patients with the low ARID5A expression were significantly shorter than those in patients with high ARID5A expression group(P<0.05).Cox regression analysis showed that the low expression and differentiation of ARID5A were independent risk factors for the prognosis of cervical cancer patients(P<0.05).Immune cell infiltration analysis showed that the expression of ARID5A in cervical cancer was positively correlated with the infiltration levels of NK cells,macrophages and B cells.Gene enrichment analysis revealed that ARID5A may be involved in humoral immune response,cytokine-cytokine receptor interaction,nuclear receptor and peroxisome proliferator-activated receptor signaling pathways.Conclusion The low ARID5A expression in cervical cancer tissues is an independent risk factor affecting the prognosis of cervical cancer
