摘 要:【目的】挖掘与马铃薯(Solanum tuberosum L.)块茎形成与发育相关的糖转运蛋白(Sugars will eventually be exported transporters),为下一步验证功能及精准运用这些基因做好准备。【方法】通过高通量转录组分析块茎发育过程中SWEETs基因的表达模式,并进一步结合蔗糖、赤霉素(GA)及光周期诱导表达分析,筛选获得块茎发育过程关键SWEET基因。【结果】通过相关诱导处理发现其中12、15、9个StSWEET基因分别在8%蔗糖、光周期、100μmol/L赤霉素处理下诱导表达。蔗糖处理下,SWEET12e、SWEET12d和SWEET2c在诱导24 h后表达量最高,而SWEET10-like的表达量随诱导时间的延长呈现波动但趋于平稳;随着短日照处理时间的延长,SWEET12e、SWEET12d和SWEET10-like的表达量显著上调;在GA处理下,SWEET12e、SWEET10-like和SWEET12d 3个基因分别在诱导后12、12和3 h表达降至低谷。只有StSWEET12e、StSWEET12d和StSWEET10-like 3个基因同时受到不同处理条件的显著诱导表达,表明其在块茎发育过程可能发挥更为关键的作用。【结论】本研究为进一步验证糖转运蛋白(SWEETs)参与马铃薯块茎发育过程提供了明确的候选基因。【Objective】 The study aimed to identify the Sugars Will Eventually be Exported Transporters(SWEETs) related to tuber formation and development in potato(Solanum tuberosum L.),in which prepare for further functional validation and accurate use of these genes.【Method】 This study analyzed SWEETs genes expression patterns during tuber development via high-throughput transcriptome analysis and combined sucrose, GA, and photoperiodic induction analysis to screen key SWEET genes for tuber development.【Result】 Through induction treatment, 12,15 and 9 StSWEET genes are induced by 8% sucrose, photoperiod, and 100 μmol/L gibberellin respectively.The study found that under sucrose treatment, SWEET12e,SWEET12d and SWEET2c had the highest expression after 24 hours of induction.The expression of SWEET10-like fluctuated but tended to be stable as induction time prolonged.As short-day treatment time prolongs, the expression of SWEET12e,SWEET12d and SWEET10-like was significantly up regulated.Under GA treatment, the expression of SWEET12e,SWEET10-like and SWEET12d genes reached the lowest at 12 hours, 12 and 3 hours after induction respectively.Only StSWEET12e,StSWEET12d and StSWEET10-like were significantly induced and expressed under different treatment conditions, suggesting they might play a more critical role in tuber development.【Conclusion】 The study provides definite candidate genes for further verifying the function of sugar transporters(SWEETs) in the potato tuber development process.
