摘 要:为探讨鳖甲煎丸(biejiajian pill,BJJP)对胰腺癌细胞增殖、凋亡和上皮-间充质转化(EMT)的干预作用.采用CCK-8、集落形成、Western blot、TUNEL划痕、Transwell和免疫荧光等实验方法检测BJJP含药血清对人正常胰腺导管上皮细胞HPNE活力的影响,对胰腺癌细胞PANC-1增殖、凋亡、迁移、侵袭、EMT以及PI3K/AKT通路蛋白表达的影响.结果显示:BJJP含药血清对HPNE细胞的活力无明显影响,具有抑制PANC-1细胞的增殖、迁移、侵袭、EMT进程的能力,促进了PANC-1细胞凋亡,抑制了PI3K/AKT通路,综上,鳖甲煎丸能抑制胰腺癌PANC-1细胞的恶性生物学行为,其机制可能与调节PI3K/AKT通路有关.To investigate the effects of BJJP on the proliferation,apoptosis,and EMT of PANC-1 pancreatic cancer cells.CCK-8,Colony formation,Western blot,TUNEL scratch,Transwell and immunofluorescence assays were used to detect the effect of BJJP-containing serum on the activity of human normal pancreatic ductal epithelial cells HPNE,and detect the effects of BJJP-containing serum on the proliferation,apoptosis,migration,invasion,EMT,and the protein expression of PI3K/AKT pathway of pancreatic cancer cells PANC-1.The results showed that BJJP-containing serum has no significant effect on the activity of HPNE cells but inhibits the proliferation,migration,invasion,EMT process and PI3K/AKT pathway,enhances the apoptosis of pancreatic cancer PANC-1 cells.Studies have shown that BJJP inhibits the malignant biological behavior of pancreatic cancer PANC-1 cells,and the mechanism may be related to the regulation of PI3K/AKT pathway.
