连翘苷对卵巢癌SKOV3细胞增殖、侵袭、凋亡及MEK/ERK信号通路的影响

摘  要:目的探讨连翘苷对卵巢癌SKOV3细胞增殖、侵袭、凋亡及丝裂原激活蛋白激酶(MEK)/细胞外信号调节激酶(ERK)信号通路的影响。方法体外培养卵巢癌SKOV3细胞和正常卵巢上皮IOSE80细胞,检测连翘苷对2种细胞增殖的抑制情况,计算半数抑制浓度(IC50);将SKOV3细胞分为阴性对照组、阳性对照组、连翘苷低剂量组、连翘苷中剂量组和连翘苷高剂量组。干预SKOV3细胞48 h后,采用流式细胞术检测细胞凋亡情况,采用Transwell法检测细胞侵袭情况,同时采用蛋白免疫印迹法检测细胞中MEK、磷酸化-MEK(p-MEK)、ERK、磷酸化-ERK(p-ERK)、上皮型E钙黏蛋白(E-cadherin)、B细胞淋巴瘤/白血病-2蛋白(Bcl-2)、Bcl-2相关X蛋白(Bax)水平。结果连翘苷浓度≥5.0μmol/L时SKOV3细胞和IOSE80细胞的增殖率均低于连翘苷浓度为2.5、0.0μmol/L时的增值率(P<0.05)。SKOV3细胞的IC50为22.59μmol/L,IOSE80细胞的IC50为73.62μmol/L。与阴性对照组比较,其余各组细胞凋亡率、E-cadherin和Bax水平升高(P<0.05),线粒体膜电位(MMP)、细胞侵袭数、p-MEK/MEK、p-ERK/ERK、Bcl-2/Bax及Bcl-2水平降低(P<0.05);与阳性对照组比较,各连翘苷剂量组细胞凋亡率、E-cadherin、Bax水平降低(P<0.05),MMP、细胞侵袭数、p-MEK/MEK、p-ERK/ERK、Bcl-2/Bax及Bcl-2水平升高(P<0.05)。结论连翘苷可以抑制SKOV3细胞增殖和侵袭,这可能与连翘苷抑制MEK/ERK信号通路,诱导SKOV3细胞凋亡有关。Objective To investigate the effect of forsythiaside on the proliferation,invasion and apoptosis of ovarian cancer SKOV3 cells,as well as its impact on the MEK/ERK signaling pathway.Methods Ovarian cancer SKOV3 cells and normal ovarian epithelial IOSE80 cells were cultured in vitro to assess the inhibitory effect of forsythiaside on cell proliferation and the half-maximal inhibitory concentration(IC 50)was calculated.SKOV3 cells were divided into a negative control group,a positive control group,a low-dose forsythiaside group,a medium-dose forsythiaside group and a high-dose forsythiaside group.After 48 h of intervention with forsythiaside in SKOV3 cells,apoptosis was detected by flow cytometry,cell invasion was detected by Transwell assay,and Western blot was used to detect MEK,phosphorylated-MEK(p-MEK),ERK,phosphorylated-ERK(p-ERK),epithelial-type E-cadherin(E-cadherin),B cell lymphoma/leukemia-2 protein(Bcl-2)and Bcl-2-related X protein(Bax)levels.Results When the concentration of forsythiaside was≥5.0μmol/L,the proliferation rates of SKOV3 cells and IOSE80 cells were lower than those at concentrations of 2.5μmol/L and 0.0μmol/L(P<0.05).The IC 50 of SKOV3 cells was 22.59μmol/L,and the IC 50 of IOSE80 cells was 73.62μmol/L.Compared with the negative control group,the apoptosis rate,E-cadherin and Bax levels increased,while mitochondrial membrane potential(MMP),the number of invading cells,p-MEK/MEK,p-ERK/ERK,Bcl-2/Bax and Bcl-2 levels decreased in the other groups(P<0.05).Compared with the positive control group,apoptosis rate,E-cadherin and Bax levels decreased(P<0.05),while MMP,the number of invading cells,p-MEK/MEK,p-ERK/ERK,Bcl-2/Bax and Bcl-2 levels increased in each forsythiaside dose groups(P<0.05).Conclusion Forsythiaside can inhibit the proliferation and invasion of SKOV3 cells,which may be related to its inhibition of the MEK/ERK signaling pathway and induction of apoptosis in SKOV3 cells.
