摘 要:目的研究m6A结合蛋白IGF2BP3在食管鳞癌中的表达水平及临床意义,并探讨其通过调节下游靶基因C-Myc的表达影响上皮间质转化(EMT)过程的机制,为发现食管鳞癌的生物标志物提供实验依据。方法分析IGF2BP3 mRNA表达与食管癌总生存期的关联,m6A结合蛋白IGF2BPs在食管鳞癌组织及正常组织的差异表达,将TCGA数据库、GEO数据集中的原始数据集进行交叉基因集综合分析,对食管鳞癌细胞组织和相邻的非肿瘤组织中IGF2BP3与C-Myc蛋白的表达水平进行检测,对m6A结合蛋白IGF2BP3与食管鳞癌患者病理特征的相关性进行分析,绘制m6A结合蛋白IGF2BPs的受试者工作特征(ROC)曲线,分析C-Myc、m6A结合蛋白IGF2BP3及EMT之间的联系。结果m6A结合蛋白IGF2BP3高表达者总生存期更短,其在食管鳞癌中差异表达最明显,且在食管鳞癌数据集中均表现高表达。食管鳞癌组织中的IGF2BP3和C-Myc相关蛋白的表达水平相较于相应的癌旁组织显著升高,m6A结合蛋白IGF2BP3与食管鳞癌患者的浸润深度呈正相关,在食管鳞癌诊断中显示出较高的诊断价值,C-Myc可能是m6A结合蛋白IGF2BP3的下游效应基因。结论m6A结合蛋白IGF2BP3在食管鳞癌组织中的表达显著升高,其表达水平的改变与食管癌患者的总生存期密切相关,具有作为食管鳞癌诊断标志物的潜力。Objective To study the expression level and clinical significance of m6A binding protein IGF2BP3 in esophageal squamous cell carcinoma,and to further explore the mechanism by which it affects the epithelial-mesenchymal transition(EMT)process by regulating the expression of its downstream target gene C-Myc,providing experimental basis for the discovery of biomarkers of esophageal squamous cell carcinoma.Methods To analyze the relationship between the expression of IGF2BP3 mRNA and the overall survival time of esophageal cancer,and the differential expression of m6A binding protein IGF2BP3 in esophageal squamous cell carcinoma and normal tissues.The original data sets in TCGA database and GEO databases were comprehensively analyzed by cross-gene set,and the expression levels of IGF2BP3 and C-Myc proteins in esophageal squamous cell carcinoma and adjacent non-tumor tissues were detected.The correlation between m6A binding protein IGF2BP3 and pathological features of esophageal squamous cell carcinoma patients was analyzed.The receiver operating characteristic(ROC)curve of m6A binding protein IGF2BP3 was drawn,and the relationship between C-Myc,m6A binding protein IGF2BP3 and EMT was analyzed.Results The overall survival time of patients with high expression of m6A binding protein IGF2BP3 is shorter.Its differential expression was the most significant in esophageal squamous cell carcinoma,and it was highly expressed in the above data sets of esophageal squamous cell carcinoma.The expression levels of IGF2BP3 and C-Myc related proteins in esophageal squamous cell carcinoma tissues were significantly higher than those in corresponding adjacent tissues.The m6A binding protein IGF2BP3 was positively correlated with the invasion depth of esophageal squamous cell carcinoma patients,which showed high diagnostic value in the diagnosis of esophageal squamous cell carcinoma.C-Myc may be a downstream effect gene of m6A binding protein IGF2BP3.Conclusion The expression of m6A binding protein IGF2BP3 is significantly increased in
