摘　　要：目的:探讨乙酰基转移酶KAT2A和Survivin蛋白乙酰化在食管癌组织中的表达情况。方法:以大理大学第一附属医院2020年3月至2022年3月确诊的60例食管癌患者为研究对象,采集所有患者的癌组织标本纳入观察组,采集距离癌组织8 cm处、无癌侵犯的正常食管上皮黏膜组织标本纳入对照组。检测并比较Survivin、KAT2A的表达水平及Survivin蛋白乙酰化程度,并对Survivin蛋白乙酰化和KAT2A进行相关性分析。结果:观察组Survivin mRNA、KAT2A mRNA的相对表达量,Survivin蛋白、KAT2A蛋白的表达量,Survivin乙酰化率均显著高于对照组,差异有统计学意义(P<0.01);食管癌Survivin乙酰化与KAT2A存在显著正相关(r=0.543);不同分期、不同分化程度以及淋巴结是否转移患者间,Survivin乙酰化、KAT2A阳性率差异有统计学意义(P<0.05)。结论:KAT2A参与了食管癌患者中Survivin蛋白的乙酰化修饰,且KAT2A的高表达可能是Survivin蛋白乙酰化修饰的前期分子事件。Objective:To investigate the expression of acetyltransferase KAT2A and the acetylation of Survivin protein in esophageal cancer tissues.Methods:A total of 60 esophageal cancer patients diagnosed at the First Affiliated Hospital of Dali University between March 2020 and March 2022 were included in this study.Cancer tissues were collected as the observation group,and normal esophageal epithelial tissues,8 cm away from the cancer site without cancer invasion,were collected as the control group.The expression levels of Survivin and KAT2A,as well as the acetylation level of Survivin protein,were measured and compared between the groups.The correlation between Survivin protein acetylation and KAT2A expression was also analyzed.Results:The relative expression levels of Survivin mRNA and KAT2A mRNA,the expression levels of Survivin protein and KAT2A protein,and the acetylation rate of Survivin in the observation group were all significantly higher than those in the control group,and the differences were statistically significant(P<0.01).There was a significant positive correlation between Survivin acetylation and KAT2A expression in esophageal cancer(r=0.543).Significant differences were found in the positive rates of Survivin acetylation and KAT2A expression among patients with different stages,different degrees of differentiation,and with or without lymph node metastasis(P<0.05).Conclusion:KAT2A is involved in the acetylation modification of Survivin protein in patients with esophageal cancer,and the high expression of KAT2A may be an early molecular event in the acetylation modification of Survivin protein.
