摘 要:本研究采用单因素试验、Plackett-Burman和响应面试验方法对米曲霉BNCC 142787的产蛋白酶条件进行优化,并探究其降解屠宰牛血液的效果。结果表明:米曲霉产蛋白酶的最佳发酵条件为发酵时间100 h,蛋白胨添加量4%,料液比1:1,此条件下的蛋白酶活力达(10221.65±53.91)U/g,较优化前升高了1.73倍。另外,通过探究该菌株对牛血液的降解效果,发现其在牛血含量33.3%的条件下,血液水解度达37%,说明该菌具有良好的血液降解效果,可用作废弃屠宰血液降解的候选菌株。In this study,the protease-producing conditions of Aspergillus oryzae BNCC 142787 were optimized by using one-way test,Plackett-Burman and response surface test methods,and the effect of degradation of slaughtered bovine blood was investigated.The results showed that the optimal fermentation conditions for protease production by Aspergillus oryzae BNCC 142787 were fermentation time of 100 h,peptone addition of 4%,and material-liquid ratio of 1:1,and the protease activity under this condition reached(10221.65±53.91)U/g,which was 1.73 times higher than that before optimization.And by exploring the degradation effect of this strain on bovine blood,it was found that its blood hydrolysis degree reached 37%under the condition of 33.3%bovine blood content,which indicated that this bacterium had good blood degradation effect and could be used as a candidate strain for the degradation of discarded slaughter blood.
